摘　　要：目的探讨胃癌组织中葡萄糖调节蛋白78(Grp78)的表达及其与胃癌病理类型、临床分期、分化程度及是否转移的关系。方法胃癌手术切除标本30例,用半定量逆转录-聚合酶链反应(RT-PCR)检测Grp78 mRNA相对表达量;免疫组织化学方法检测Grp78蛋白表达情况。结果半定量RT-PCR显示胃癌组织与正常组织中都有Grp78表达,胃癌组织为1．02±0．16,正常组织为0．62±0．14,差异具有统计学意义。其表达程度与肿瘤大小成正比、随肿瘤分化程度降低而升高,Ⅲ、Ⅳ期患者显著高于I、Ⅱ期,有淋巴结转移者表达程度显著高于无淋巴结转移者,与患者年龄及性别无关。免疫组织化学显示:胃癌组织大多数表达在++～+++,正常组织大多表达在-～++。 Grp78蛋白表达程度与肿瘤大小成正比、随肿瘤分化程度降低而升高,Ⅲ、Ⅳ期患者显著高于I、Ⅱ期,有淋巴结转移者表达程度显著高于无淋巴结转移者。结论胃癌组织的Grp78表达呈增高趋势,参与了胃癌的发展过程,并与胃癌部分生物学行为有关。Objective To study the expression of Grp78 in human gastric cancer specimens and its clinical significance. Methods Thirty cases of human gastric cancer specimens were collected from the patients who received operation for gastric cancer from May 2001 to August 2003. Semi-quantitative RT- PCR assay and immunohistochemistry were used to detect the Grp78mRNA and Grp78 protein expression respectively. Results Semi-quantitative RT-PCR revealed that the expression of Grp78 mRNA in the gastric cancer specimens and normal gastric specimens was 1.02 ＋ 0.16 and （0.62 ＋ 0.14） respectively （P〈 0.01 ）. Immunohistochemistry showed the level of Grp78 protein expression of the cancer specimens（average at q-st-Hot ） was significantly higher than that of the normal gastric specimens （average at - - ＋＋ ）. The levels of the Grp78 mRNA and protein expression were related with the size, the clinical stage, the degree of malignancy of caner, and node metastasis, but not related with the patient＇s sex and age. Conclusion The levels of Grp78 were elevated in the human gastric cancer specimens. Grp78 may play some roles in the development of gastric cancer, and was related with partial biological behaviors.
