摘 要:目的探讨外源性p14ARF基因导入人胰腺癌PC-3细胞株后,细胞侵袭力的变化。方法用脂质体介导法将外源性p14ARF基因导入人胰腺癌PC-3细胞株后,用细胞穿透Matrigel 胶法比较转染前后细胞侵袭力的改变,并用逆转录-聚合酶链反应(RT-PCR)法检测与侵袭转移相关基质金属蛋白酶(MMP)2和MMP9表达。结果转染p14ARF基因后的PC-3细胞穿过Ma- trigel胶的能力减弱,48 h后平均穿膜细胞数从(92±12)个减少到(28±5)个,而且MMP9 mRNA 表达下降。结论 p14ARF基因能抑制胰腺癌PC-3细胞的侵袭能力。Objective To observe the change of invasion of the pancreatic carcinoma cell line PC- 3 after transfected with p14ARF gene. Methods The model of pancreatic carcinoma cell line PC-3 transfected with p14ARF gene was established by lipofectin reagent protocol. The alteration of cellular migration property of PC-3, PC-3/vec and PC-3/p14 cells was evaluated by cell migration through a Matrigel model basement membrane. The expression of MMP2 and MMP9 was detected by RT-PCR. Results The average number of transwell cells with p14ARF in 48 h was reduced from 92 ± 12 to 28 ± 5. The expression of MMP9 was reduced in PC-3 cells with p14ARF. Conclusion p14ARF gene can inhibit the invasion ability of PC-3 cell line.
