摘  要:目的探讨可耐受氟他胺的LNCaP细胞亚株-LNCaP-flu耐受氟他胺的机制。方法抽提正常LNCaP细胞和LNCaP-flu细胞中的mRNA,琼脂糖凝胶电泳证明A260/A280>2.0,Su- perscriptⅡ逆转录酶行逆转录,加入Cy5和Cy3,制备cDNA探针,使用16 184点位cDNA基因芯片观察两者细胞雄激素受体相关基因表达差异情况。并使用逆转录聚合酶链反应验证基因芯片结果。结果 Trizol法提取细胞mRNA合格,从16 184个点位中筛选出差异表达基因共2 428个,占 15.0%;其中1 194条下调,1 234条上调。其中与雄激素受体功能明显相关的共有4条基因,分别为NCOR1、NCOA2、NCOA4和DDC。此4基因在氟他胺短时作用的LNCaP细胞中均无表达变化。结论雄激素受体相关基因,尤其是4种雄激素受体共调节物,在LNCaP细胞的氟他胺耐受性的产生过程中可能起到一定作用。Objective To explore the molecular mechanism underlying LNCaP-flu cell line's flutamide insensitivity. Methods mRNA was extracted from LNCaP cell, hydroxyflutamide treated LNCaP ceil and LNCaP-flu cell. An agarose gel electrophoresis proved that A z6o/A 2s0 〉 2.0. The mRNA extracted from all groups was reversely transcribed with SuperscriptlI enzyme to make Cy5 and Cy3 labeled cDNA probes. The differential expression of androgen receptor related genes in all groups was detected by 16184 points' cDNA gene chips. Results The mRNA extracted by Trizol method was well qualified for genechips analysis. In all the 16184 genes investigated, 15.0% of them, 2428 genes, weree expressed differentially. Among them, 1194 were down-regulated and 1234 up-regulated. Four of them, viz. NCOR1, NCOA2, NCOA4 and DDC, were androgen receptor related genes. All these 4 genes did not change much after ephemeral flutamide treatment in LNCaP cells. Conclusion Androgen receptor related genes, especially the 4 androgen receptor coregulators, may play a role in the gaining of flutamide insensitivity of LNCaP cells.
