摘 要:目的探讨紫龙金单独应用或者与磷酸雌二醇氮芥(EMP)联合应用对小鼠前列腺癌的体内抑瘤作用。方法建立TRAMP-C2可移植性前列腺癌动物模型,将转基因小鼠前列腺癌细胞株TRAMP-C2接种入C57小鼠,经传代至生物学特性趋于稳定,观察单纯应用紫龙金及与EMP联合应用对模型肿瘤的体内抑制作用。结果TRAMP-C2接种入C57小鼠,经体内传代6代后肿瘤生长基本稳定,适宜药物实验。治疗量紫龙金对小鼠前列腺癌的生长具有明显抑制作用,抑瘤率为28.08%,治疗量EMP抑瘤率为41.92%,2药低剂量联合应用抑瘤率达55.00%。紫龙金组小鼠实验终末体重与对照组比较无明显差异,但高于阳性对照EMP组和低剂量联合组(P<0.05)。结论紫龙金单独应用对TRAMP-C2可移植性前列腺癌具有一定的抑制生长作用,与EMP联合应用则可明显增强EMP的体内抑瘤效果,具有增效减毒作用。Objective To investigate the in vivo antitumor effects of Chinese herbal medicine Zilongjin alone or combined with estramustine phosphate (EMP) on murine prostate cancer cell TRAMP-C2. Methods A TRAMP-C2 animal model for prostate cancer research were established. TRAMP-C2 cells were cultured and injected subcutaneously into male C57BL/6 mice to establish the murine model. Zilongjin alone or combined with EMP was administered orally and the anti-tumor effects were observed. Results The TRAMP-C2 model was established successfully,and suitable for evaluation of therapeutic strategies. The in vivo study showed that the growth of the tumors was significantly inhibited by Zilongjin. The inhibition rate of Zilongjin was 28.08% ,while those of EMP and EMP combined with Zilongjin at low dose were 41.92% and 55.00% ,respectively. At the end of the experiment, the body weigh of Zilongjin group was not significantly different from that of control group,but heavier than that of EMP group and EMP plus Zilongjin at low dose group (P 〈 0.05). Conclusions Zilongjin alone can inhibit the growth of TRAMP-C2 in vivo. When combined with EMP, Zilongjin can markedly enhance the antitumor effects and decrease the side effects.
