肠胃清口服液抑制结肠癌细胞LS-174T侵袭转移的体外研究

摘　　要：[目的]研究肠胃清口服液在体外抑制结肠癌细胞侵袭转移的作用。[方法]SD大鼠肠胃清连续灌胃给药3 d后断头取血,分离血清,MTT法检测结肠癌细胞LS-174T与人工基底膜的黏附能力;划痕试验测定细胞的运动能力;浸润杯试验法研究细胞的浸润能力。[结果]肠胃清药物血清可以有效地抑制肿瘤细胞黏附能力[半数有效浓度(EC50)为90μl//ml];抑制LS-174T癌细胞移动(EC50为100μl/ml);肠胃清药物血清处理过的结肠癌细胞浸润穿过Matrigel膜的细胞数要比对照的细胞数明显减少,100μl/ml肠胃清药物血清对细胞浸润的抑制率达到56%。[结论]肠胃清口服液可以通过抑制结肠癌细胞的黏附、运动、浸润能力,抑制肿瘤的侵袭转移。[Objective] To explore the effects of Changweiqing oral liquid （COL） on the invasion and metastasis of colon cancer cell in vitro. [Methods] Melanoma cells B16BL6 were injected into C57BL/6 mice by caudal vein, then the mice were randomly divided into five groups： the control group, Dacarbazine group, low dose of COL group（12. 5 g/kg）, moderate dose group（25 g/kg） and high dose group（50 g/kg）. The mice＇s weight, behaviour and metastasis points were obversed . The serum was obtained from SD rats which had been fed with COL twice a day for three days. The adhesiveness, migration and invasion were measured by MTT assay, wound healing assay and transwell assay respectively. [Results] COL had no effect on the weight and bahaviour of mice. COL significantly inhibited metastasis in lung in the moderate and high dose groups. The serum containing of COL could inhibit adhesiveness and migration of colon cancer cell line LS-174T , the EC50 of which were 90 μl/ml, 100 μl/ml respectively. Compared with that of control group, the number of cells that infiltrated and went straight through matrigel membrane significantly decreased after the cells had been dealt with the serum. The inhibition rate of invation was 56 % when the serum concentration was 100μl/ml. [Conclusion]COL can restrain invasion and metastasis of colon cancer cells by inhibiting it＇s ability of adhesiveness, migration and invasion.
