摘 要:应用大鼠肝脏隔离灌注(ILP)技术.经门静脉灌注大剂量5-Fu观察肝脏酶学变化。结果ILP平衡液组:ILP 5-Fu 150mg/kg组,ILP 5-Fu 250mg/kg组,术后琥珀酸脱氨酶(SDH)、镁—三磷酸腺苷酶(Mg^(2+)—ATP_(ase)、5’—核苷酸酶(5’—NT)、胆碱酯酶(ChE)活性下降,糖原反应(PAS)减弱,乳酸脱氢酶(LDH)和酸性磷酸酶(AcP)活性增强。术后 3天有所恢复,7天恢复正常。ILP 5-Fu 500mg/kg 组酶学变化更为显著,术后 3天不恢复,与肝脏超微结构改变相符,表明在药物最大耐受剂量(MTD)范围内ILP区域化疗时对肝细胞的代谢功能影响轻微,超MTD的5-Fu明显影响肝细胞的代谢功能。Histochemical changes of the liver were observed using ILP technique with large dose 5-Fu in rats. The results confirmed that the activities of SDH,Mg2+-AT-Pase,5-NT ChE and PAS were decreased and the activities of LDH and ACP increased in groups of ILP without 5-Fu and ILP with 5-Fu(150 and 250mg/kg)postoperatively. The enzymatic activities began to return on the third postoperative day. The enzymatic activities in group of ILP with 500mg/kg 5-Fu had more remarkable changes and did not return to normal on the third postoperative day,but they return to normal on the sev-enth postoperative day. The changes of enzymatic activities were in agreement with those of hepatic ultrastructure. The results demonstrated that the regional chemotherapy applying ILP within MTD of 5-Fu slightly affected the metabolic functions of liver. If 5-Fu doses should exceed MTD,the metabolic functions of hepatocytes were obviously in-jured
