摘  要:利用一次腹腔注射的染毒方法观察了苯并(a)在(BaP)对小鼠脾淋巴细胞钙稳态的影响,并初步探讨了其与免疫抑制的关系。结果表明,LACA雌性小鼠一次腹腔注射BaP200mg/kg后第3天和第6天,胸腺和脾脏明显萎缩,与对照组比其相对重量分别下降46.0%~49.2%和28.7%~33.0%;全脾细胞数明显减少,同时刀豆素A(ConA)刺激的淋巴细胞增殖反应也受到明显抑制,抑制率为21.1%~27.5%;淋巴细胞内游离钙浓度([Ca ̄(2+)]i)在染毒后第3天明显升高,第6天明显下降;不同剂量BaP染毒后第3天淋巴细胞内[Ca ̄(2+)]i与染毒剂量呈明显正相关(r=0.9853,P<0.01);第6天[Ca ̄(2+)]i与染毒剂量呈明显负相关(r=-0.9473,P<0.01)。此外,还观察到钙离子载体A_(23187)使正常小鼠脾淋巴细胞[Ca ̄(2+)]i明显升高时,淋巴细胞增殖反应受到明显抑制。结果提示,BaP可明显影响小鼠脾淋巴细胞的钙稳态,而钙稳态的失衡与BaP引起的免疫抑制可能有密切的关系。Femal LACA mice were exposed to BaP by a single intraperitoneal injection to investi-gate the effect of BaP on calcium hemeostasis of splenic lymphocyte and the relationship between thiseffect and immunossuppression induced by BaP.A significant decrease in relative weights of the thymus(46.0%~49.2%)and spleen(28.7%~33.5%)was observed in mice exposed to 200 mg/kg BaP 3 and6 days after exposure. At the same dose,the number of splenocytes was significantly decreased (24%~81%)and the splenic lymphocyte proliferation by ConA was also inhibited (21.1%~27.5%)signifi-cantly in exposed mice 3,6,and 9 days later. On day 3 and day 6 after injection the intracellular free cal-cium concentration of lymphocyte was increased (r=20.985 3,P<0. 01)and decreased (r=-0.947 3,P<0. 05)in a dose-dependent manner.The intracellular free calcium level of splenic lymphocytes in nor-mal mice was also elevated by calcium ionophore A_(23187),whereas the proliferation by ConA was de-creased significantly,The results demonstrated that BaP caused disturbance of calcium homeostasis,which may relate with immunosuppression induced by BaP.
