摘  要:目的研究抗癌中药莪术油、华蟾素、龙葵碱与肝癌细胞株SMMC7721、胃癌细胞株NKM45作用后的ATP水平,探讨药物对瘤细胞增殖活性的抑制作用。方法应用三磷酸腺苷生物发光法(ATP-BLA法)测定莪术油、华蟾素和龙葵碱与肝、胃癌细胞作用后的ATP含量,并与5-氟脲嘧啶(5-FU)的作用进行比较。结果莪术油、华蟾素、龙葵碱与SMMC7721、NKM45作用后的ATP含量明显下降(P<0.05~P<0.01);与5-FU作用后的ATP含量比较,无明显差异(P>0.05)。结论应用ATP-BLA研究抗癌中药对肿瘤细胞增殖活性的抑制作用,对于筛选抗肿瘤药物具有广阔的应用前景。Objective To explore of inhibiting proliferation activity of hepatoma cell (SMMC7721), gastric carcinoma cell (NKM45) by three anti-tumor Chinese materia medica such as curcuma aramatica oil etc. Methods ATP-bioluminescence assay (ATP-BLA) was used to determine concentration of ATP when SMMC7721 and NKM45 were acted on by cureuma aramatica oil ,cinobufacin and solanine. The concentration of ATP was also compared with of tumor cells that were acted on by 5-Fluorouracil (5-FU). Results The concentration of ATP was decreased significantly when the tumor cells were acted on by curcuma aramatica oil,cinobufacin and solanine (P〈0.05~P〈0.01). There was no significant difference when they were compared with of the tumor cells that were acted on by 5- FU (P〉0.05). Conclusion ATP-BLA is a reliable assay for inhibiting propagation activity of tumor cells by anti-tumor Chinese material medica. There are vast prospect in screening drugs of anti-tumor by the use of ATP-BLA.
