改良染料排斥法体外药物敏感试验预测急性白血病化疗的反应

摘　　要：使用改良染料排斥法测定１７例２０人次的白血病患者对常用的９种化疗药物体外的敏感性，其中急性白血病１４人（初治７人，复发３人，难治４人），ＦＡＢ分型ＡＮＬＬ１０人（Ｍ１１人，Ｍ２５人，Ｍ５３人，Ｍ６１人），ＡＬＬ４人（Ｌ２３人，Ｌ３１人），ＭＤＳ－ＲＡＥＢＴ１人ＣＭＬ急变２人，体外药敏结果显示：体外药敏实验与体内疗效总符合率为５９．２％，阳性符合率６５．８２％，阴性符合率４８．９１％。初步表明该法对指导临床白血病个体化治疗，选用有效的化疗药物有一定的应用价值。本方法还具有准确率高、操作简便、所需标本少和实验时间短的优点。The cytotoxicities of cytosine arabinoside,vincristin 'daunorubicin,aclacinomycin,mifoxantrone,etoposide,mitoxantrone,harringtonine,cycfophosphamide to leukemia cellwere measured with novel dye exclusion mothod in 17 paticnts with leukemia to observe the rela-tionship between chemosensitivity and efficacy in vivo. Leuken1ia cells were incubated with anti-tumor agents in micro-well culture system for 48 hours and the mean cytotoxicity of single drugwas used as in vitro sensitivity index. In vivo sensitivity was defined as complete remission or de-scent of more than 5 0%blasts in bonc marrovv after a course of chemtherapy.The resultshowed that the positive and negative coinciderit rates between chemosensitivity and efficacv invivo were 59.2%,65.82%and 48.91% respectively.
