摘 要:目的研究川芎嗪对哮喘大鼠肺组织中核因子-κB(NF-κB)表达的影响及其抗炎作用。方法将SD大鼠随机分为4组:对照组、模型组、激素组和川芎嗪组。用卵蛋白复制大鼠哮喘模型;免疫组织化学技术观察气道上皮细胞NF-κB的表达;行支气管肺泡灌洗液(BALF)中嗜酸性粒细胞(Eos)计数及细胞分类计数。结果(1)4组NF-κB细胞核阳性表达的细胞占气道上皮细胞的百分比分别为(12.1±2.4)%、(26.8±3.6)%、(14.6±1.9)%和(17.3±2.6)%。模型组明显高于对照组(P<0.001);川芎嗪组明显低于模型组(P<0.01),且与激素组无显著性差异(P>0.05)。(2)川芎嗪组BALF中Eos的数目及Eos与淋巴细胞的比例均低于模型组(P<0.001),且其作用与激素组无显著性差异(P>0.05)。结论川芎嗪可以有效阻断肺组织中NF-κB的激活,减轻气道炎症。Objective To explore the effects of ligustrazine on nuclear factor-kappe B (NF-κB) expression and airway inflammation in a rat model of bronchial asthma. Methods Forty male SD rats were randomly divided into 4 groups: normal control group, asthmatic model group, desamethasone group and ligustrazine group. The asthmatic model was established by repeated inhalation of ovalbulium. The expression of NF-κB in the lung tissues was ob- served by means of immunohistochemical staining. The eosinophil (Eos) amounts and differential leukocytes counts in bronchoalveolar lavage fluid (BALF) were counted . Results ( 1 ) In the four groups, the nuclear expression of NF-κB in epithelial cells were ( 12.1 ± 2.4) %, (26.8 ± 3.6) %, ( 14.6 ± 1.9) %, ( 17.3 ± 2.6) % respectively. There was a significant increase in expression of NF-κB in the lung tissues in asthmatic model group compared with that in normal control group( P 〈 0. 001 ). The expression of NF-κB in ligustrazine group was much lower than that in asthmatic model group( P 〈 0. 01 ), There were no statistical significant difference between ligustrazine group and desamethasone group( P 〉 0.05). (2) The Eos amounts in BALF in lignstrazine group was much lower than that in asthmatic model group( P 〈 0. 001 ), There were no statistical significant difference between ligustrazine group and desamethasone group( P 〉 0. 05). There was a significant decrease in lymphocytes and eosinophils in BALF in ligustrazine group compared with asthmatic model group( P 〈 0. 001 ). Conclusion Ligustrazine can inhabit the expression of NF-κB and decrease airway inflammation in asthmatic rats.
