摘 要:目的探讨细胞外基质降解在猪后肢侧支血管生长过程中的作用。方法成年家猪6只,单侧股动脉结扎,诱导侧支血管生长,用共聚焦免疫荧光术研究基质金属蛋白质酶-2(MMP-2)和组织Ⅰ型基质金属蛋白质抑物(TIMP-1)在侧支血管的表达。同时用VanGieson弹性染色观察了血管组织中弹性成分的变化。结果在正常血管,血管的管腔侧可以观察到一层完整的内弹性膜,而在生长的侧支血管,内弹性膜发生降解,片断化,可观察到有新内膜形成;在正常血管MMP-2表达水平很低,而在生长血管MMP-2高水平表达,尤其在新内膜区域,是正常血管的9倍。TIMP-1的表达在内膜为低水平,但在中膜是高水平的表达。结论该研究首次报道猪后肢侧支血管生长过程中细胞外基质的降解十分活跃,对侧支血管的生长具有重要的作用。[Objective] To investigate the role of extracellular proteolysis during collateral vessel growth in pig hind limb. [Methods] Six adult pigs were used. The surgery for unilateral ligafion of femoral artery was performed to induce collateral vessel growth. Expression of MMP-2 and TIMP-1 was detected by confocal microscopy. In addition, Histology was examined using van Gieson staining. [Results] In normal vessels, an entire layer of internal elastica lamina (IEL) was observed, however, in growing collateral vessels, IEL was disrupted, neointima was evident. In normal vessels, expression of MMP-2 was very low, but highly up-regulated in growing collateral vessels, especially in the neointima, about 9 times higher than that in normal vessels. [Conclusion] This data for the first time demonstrate that extracellular proteolysis is very active during collateral vessel growth in pig hind limb, implying its importance for enlargement of collateral vessels.
