Ang-2在胃癌组织中表达与胃癌新生血管形成的关系

摘　　要：目的研究Ang-2(angiopoietin-2)在胃癌组织中的表达与胃癌新生血管形成的关系,并探讨其临床意义。方法用Western Blot法检测Ang-2蛋白在10例胃癌组织及10例正常胃组织中的表达;用免疫组化法检测53例胃癌组织中Ang-2蛋白的表达。结果①Ang-2蛋白在正常胃组织中不表达,胃癌组织中呈阳性表达;②Ang-2蛋白在胃癌的不同临床病理分期中表达强度不同,且随病程进展表达呈增强趋势,各组间差异有统计学意义;③胃癌组织中微血管密度(13±2/HP)和新生血管的数量(8±3/HP)明显高于正常胃组织(4±1/HP,阴性)。结论Ang-2参与了胃癌组织新生血管的形成及维持,该蛋白可以作为监测胃癌进展及评价预后的指标之一。Objective To study the expression of angiopoietin-2 in gastric carcinoma tissue and its relation-ship with tumor angiogenesis, and to explore its clinical significance. Methods Immunohisto- chemical method was used to detect the expression of Ang-2 protein in 53 samples of gastric cancer tissue, and western blot technique was used to detect the expression of Ang-2 protein in 10 samples of gastric carcinoma tissues and 10 normal gastric tissues. Results Ang-2 was expressed in tumor tissue, while normal tissue was unexpressed. The expression intensity of Ang-2 was strongly associated with stages of tumor. The microvessel density（MVD, 13±2/HP） and the average number of informed vessels （IMV, 8±3/HP） of gastric carcinoma tissues were obviously higher than those of normal tissues（4±1/HP, negative） Conclusion Ang-2 plays an important role in the progress of angiogenesis and tumor growth, it can be used to evaluate the growth of gastric carcinoma and predict prognosis in gastric cancer.
