摘 要:利用有机溶剂分级沉淀法从榆树叶中分离出超氧化物歧化酶(SOD),研究分离纯化的最佳工艺条件,并对其性质进行探讨.结果表明:当榆树叶质量与磷酸缓冲液体积之比为1∶3、磷酸缓冲液pH值为7.8、提取时间为1.5 h、氯仿-乙醇混合物量为提取液的0.25倍、丙酮量为粗酶液的1倍时,分离效果最佳.温度0~60 ℃,pH值在4~9范围内酶的活性稳定.该酶对KCN和H2O2敏感,表明酶的类型为Cu·Zn-SOD.研究结果为SOD提供了新的酶源.The superoxide dismutase from elm leaves has been isolated and purified by organic solvent fractionation by precipitation. Optimum technological condition of isolation and purification and its property were studied. The results showed that the optimal isolating condition were that the ratio of the elm leaves to the phosphoric acid buffer solution was 1:3 (m/V), the phosphoric acid buffer solution pH value was 7.8, the extractive time was 1.5 h. the chloroform-ethyl alcohol mixed solution amount was 0.25 time of extracts, the acetone amount was 1.0 time of coarsely enzyme fluid, the enzymatic activity is stable within the temperature 0-60 ℃ and pH 4-9, and the enzyme is sensitive to KCN and H2O2, the facts indicated that the enzyme is Cu·Zn-SOD. This study supplied new enzyme resource for superoxide dismutase.
