摘  要:建立了一种雨生红球藻中虾青素酯皂化的反相高效液相色谱方法.采用二极管阵列检测器(DAD)在476 nm处进行检测,虾青素在15 min内得到了较好的分离.虾青素的含量在1~15 mg/L范围内与峰面积具有良好的线性关系,相关系数为0.999 3;方法的检出限为0.049 mg/L,RSD为3.27%,及回收率为100.7%.应用本法测定出雨生红球藻中虾青素的总含量为9.79~24.70 mg/L.A reversed-phase high performance liquid chromatographic (RP-HPLC) method was developed for the separation of astaxanthin ester that have been saponified from Haematococcus pluvialis. Chromatographic separation was performed on Hypersil ODS column (5μm, 4.6 × 250 mm, Germany) at room temperature with an eluent of the mixlure of acetonitrile and methanol (75:25,V/V) al a flow rate of 1. 0 mL/min. The eluate was monitored by a diode array detector at 476 nm. Separation and identification of astaxanthin was completed within 15 min. The detection limit and the linear range of astaxanthin were 0. 049 mg/L and 1~15 mg/L (r=0. 9993),respectively. The pre (RSD) was 3.27%, and average recovery was 100.7%.
