摘 要:目的:探讨HER2、NF-kB、VEGF-C及其受体VEGFR-3在乳腺癌组织中的表达及相互关系。方法:应用免疫组化方法检测40例人乳腺癌组织和10例癌旁组织HER2、ER、NF-kB、VEGF-C及VEGFR-3的表达情况。结果:乳腺癌组织和癌旁组织NF-kB的阳性率分别为45.0%(18/40)和30.0%(3/10),差异无显著性(P>0.05),NF-kB与HER2蛋白的表达呈正相关(r=0.442,P=0.004)。Ⅲ级乳腺癌的NF-kB表达显著增强(P=0.018)。VEGF-C和VEGFR-3的表达显著增强,阳性率分别为70.0%(28/40)和57.5%(23/40),两者呈正相关(r=0.761);NF-kB和HER2与VEGF-C呈正相关(r值分别为0.373和0.342)。淋巴结转移组VEGF-C和VEGFR-3的阳性率分别等于90.0%和82.4%,明显高于无淋巴结转移组(56.5%和39.1%)。VEGFR-3阳性脉管数平均为8.44±2.58,明显高于无淋巴结转移组(5.12±3.75)。结论:NF-kB与HER2的表达为正相关,NF-kB和HER2蛋白与VEGF-C的表达呈正相关。Objective: To investigate the protein expression of nuclear transcription factor NF-KB, VEGF-C and its receptor VEGFR-3 in breast cancers, and the correlations among NF-KB, VEGF-C, VEGFR-3 and histological grading and nodal status. Methods: Forty samples of human breast cancer tissue and adjacent non-neoplastic breast tissue were examined for NF-KB, VEGF-C,VEGFR-3, as well as HER2 and ER by immunohistochemistry. Results: The expressions of NF-KB in breast cancer tissue and adjacent non-neoplastic breast tissue were 45.0%(18/40)and 30.0% (3/10, respectively. The expression of NF-KB in breast cancer has no significant difference compared to that in adjacent nonneoplastic breast tissue. NF-KB and HER2 expressions were positively correlated (r=0.442, P=0.004). The expression of NF-KB in the group of grade 111 was increased compared to the other two groups (P=-0.018). The positive rate of VEGF-C and VEGFR-3 in breast cancer were 70.0%(28/40)and 57.5%(23/ 40)respectively. The expression of VEGF-C and VEGFR-3 in breast cancer were increased in breast cancer (P〈0.05), as compared with that in non-neoplastic breast tissue. The expression of VEGF-C correlated significantly with VEGFR3 (r=0.761). Both NF-KB and HER2 were correlated with VEGF-C (r value were 0.373 and 0.342 respectively). The positive rate of VEGF-C and of VEGFR-3 were 90.0% and 82.4% respectively in the lymph-node metastasis group, which was significantly higher thanthat of non-lymph node metastasis group (56.5% and 39.1% respectively). The number of VEGFR-3 positive vessels in lymphnode metastasis group (8.44±2.58) was significantly higher compared to the nonlymph node metastasis group (5.12±3.75). Conclusion: NF-KB is activated abnormally in part of the breast cancers. The expression of the NF-KB and of the HER2 protein positively correlate; both NF-KB and HER2 expression positively correlate with VEGF-C.
