摘 要:目的探讨KAI1基因对MHCC97-H肝癌细胞粘附力的影响,从而推测KAI1基因影响MHCC97-H肝癌细胞侵袭转移的机制。方法采用微管吸吮技术研究我们前已转染人类KAI1全长正、反义结构基因的人肝癌MHCC97-H细胞粘附力。结果在纤维连接蛋白(fibronectin,FN)裱衬表面,利用微管吸吮系统测得的各组肝癌细胞的粘附力分别为MHCC97-H-S组(678·4±101·8),MHCC97-H-AS组(1083·6±113·8),MHCC97-H-pCI组(853·0±105·4)及MHCC97-H亲本细胞组(834·6±130·5)(单位为10-10N,细胞数为25)。与MHCC97-H亲本细胞组比较,MHCC97-H-S组细胞对FN的粘附力显著下降(P<0·01),MHCC97-H-AS组细胞对FN的粘附力显著增加(P<0·01),而MHCC97-H-pCI组细胞对FN的粘附力无明显变化(P>0·05)。结论KAI1基因可能降低肝癌细胞对细胞外基质FN的粘附力,从而抑制转移的初始步骤,进而抑制MHCC97-H肝癌细胞的侵袭和转移。Objective To explore the effects of metastasis suppressor gene KAII on adhesion forces of MHCC97-H hepatocellular carcinoma cells with high metastatic potential, so as to find the possible mechanism. Methods The adhesion forces of MHCC97-H cells with high metastatic potential transfected with sense or antisense KAI1 expression plasmid were measured by means of micropipette aspiration technique. Results On FN coated surfaces, the cell adhesion forces were quantitatively described by means of micropipette aspiration technique. The forces of the MHCC97-H cells were significantly higher after transfected with antisense KAI1 expression plasmid [ ( 1 083.6 ±113.8 ) 10^-10 N ] (P 〈 0. 01 ), lower after transfected with sense KAI1 expression plasmid [ (678.4± 101.8) 10^-10 N] (P 〈 0. 01 ), and of no significant difference after transfected with its control vector pCI-neo without KAI1 gene [ (853.0 ± 105.4) 10^-10 N] (P 〉0. 05), as compared with their paternal MHCC97-H cells [ (834.6 ± 130. 5) 10^-10 N ]. Conclusion The metastasis suppressor gene KAI1 may decrease the cell adhesion forces to extracellular matrix to inhibit metastasis from first step, furthermore inhibit the invasion and metastasis of MHCC97-H cells.
