摘  要:目的:探讨全反式维甲酸(All trans retinoic acid,ATRA)对人子宫内膜腺癌细胞株(RL95-2)的增殖抑制作用。方法:用不同浓度ATRA处理体外培养的RL95-2,分别在处理后不同的时间点,以MTT法检测ATRA对RL95-2增殖活性的影响;用流式细胞术检测经ATRA处理后的RL95-2细胞周期的变化;免疫组化方法检测细胞周期素A(Cyclin A)的改变;扫描电镜和透射电镜观察ATRA处理后的细胞形态变化。结果:ATRA可抑制RL95-2的增殖,且这种作用呈时间及浓度依赖性。FCM分析发现RL95-2细胞增殖被ATRA阻滞于G0/G1期。电镜观察到用药后细胞恶性表型消失。免疫组化发现AT- RA处理后48h,Cyclin A蛋白表达降低。结论:ATRA对子宫内膜癌细胞系RL95-2的生长有抑制作用,且这种抑制作用与 Cyclin A蛋白表达水平降低有关。Objective: To evaluate the action of all trans retinoic acid on human endometrial adenocarcinoma cell line RL95 -2. Methods : The human endometrial adenocarcinoma cells ( RL95 - 2 ) cultured in vitro were treated with various concentrations of all trans retinoic acid(ATRA). At specific time points after treatment with ATRA, the effect of ATRA on the proliferation of RL95 -2 cells was determined by MTT method, and Cyclin A was detected by immunohistochemistry. The morphological change of RL95 - 2 cells was observed by scanning electron microscope and transmission electron microscope. Results : A time - dependent and dose - dependent proliferation inhibition was demonstrated in RL95 -2, and ATRA induced a G0/G1 cell cycle arrest. The malignant morphological characteristic of RL95 - 2 cells disappeared after treatment by ATRA. ATRA could down - regulate the expression of Cyclin A. Conclusion : The proliferation inhibition effect of ATRA on RL95 - 2 cells cultured in vitro appears to be due to the down - regulation of Cyclin A. The results provide evidence for the potential effect of ATRA in the prevention and treatment of human endometrial adenocarcinoma.
