转小鼠TNFB基因大鼠脑缺血时细胞凋亡、c-JUN、p-JUN与p-JNK表达的研究

摘　　要：目的:探讨肿瘤坏死因子B(TNFB)过度表达时对细胞凋亡、c-JUN、p-JUN与p-JNK表达的影响。方法:采用线栓法制作大鼠大脑中动脉闭塞模型,用TNFB、TUNEL、c-JUN、p-JUN与p-JNK免疫组化染色观察SD大鼠与转小鼠TNFB基因大鼠缺血1h再灌注24h时的细胞凋亡、c-JUN、p-JUN与p-JNK的表达。结果:与SD大鼠相比,转小鼠TNFB基因大鼠缺血1h再灌注24h后,脑组织凋亡细胞显著增多,c-JUN、p-JUN与p-JNK过度表达显著,以上指标变化均显著高于SD大鼠。结论:TNFB的过度表达加剧缺血性脑损伤,其机制可能为c-JUN、p-JUN与p-JNK的过度表达。Objective：To explore the effects of over expression of tumor necrosis factor-α（TNF-α） in cerebral ischemia tissues on apoptosis, expression of c-JUN, phospho-c-jun （p-JUN） and phospho-c-jun N-terminal kinase （p-JNK）. Methods ：The right middle cerebral artery in Sprague-Dawley rats and murine TNFa transgenic rats was occluded by suture occlusion technique. Apoptosis, protein expression of c-JUN, p-JUN and p-JNK were examined when the cerebral ischemia tissues of Sprague-Dawley rats and murine TNFa transgenic rats subjected to 1 hour occlusion and 24 hours reperfusion by immunohistochemistry of terminal deoxynucleotidyl transferase（TdT）-mediate biotinylated UTP nick end labeling （TUNEL）technique, c-JUN, p- JUN and p-JNK staining. Results： Excluding the increased level of apoptosis, c-JUN, p-JUN and p-JNK overexpressed significantly in the murine TNFa transgenic rats subjected to 1 hour occlusion and 24 hours reperfusion , which were higher than those in the Sprague-Dawley rats. Conclusion： Overexpression of TNFa gen strongly effects on cerebral ischemia injury, which is possibly related with c-JUN, p-JUN and p-JNK overexpression.
