摘  要:目的:研究一氧化氮(NO)和内皮素-1(ET-1)在大鼠肢体缺血/再灌注(L I/R)后脑氧化损伤中的作用,探讨NO/ET-1平衡关系的变化对脑损伤的影响。方法:在大鼠L I/R损伤模型上,观察血浆NO,ET-1,MDA,XOD,SOD,LDH,CK及脑组织tNOS,iNOS,cNOS,NO,ET-1,MDA,XOD,M PO,SOD,W/D的变化。结果:L I/R后血浆LDH,CK,MDA,XOD和脑组织MDA,XOD,M PO,W/D较对照组升高,自由基清除酶SOD活性下降,在再灌注4h脑组织损伤最为严重。再灌后,脑组织tNOS和iNOS活性升高,而cNOS活性降低,血浆及脑组织中NO,ET-1增加,NO/ET-1比值降低,在再灌注4h降低最为明显。结论:大鼠L I/R后脑损伤的发生可能与机体自由基产生增多,清除酶活性下降以及NO/ET-1失平衡有关。Objective.. To study the roles of nitric oxide and ET-1 in brain oxidation injury afte, hind limbs ischemia-reperfusion in rats and to investigate the effect of NO/ET-1 balance on brain injury. Methods: The changes of NO,ET-1,MDA,XOD,SOD,LDH,CK in plasma and tNOS,iNOS,cNOS,NO ET-1, MDA, XOD, MPO, SOD, W/D in brain tissue were observed on the rat model of the hind limb ischemia-reperfusion (LIR). Results; After rats hind limbs suffered ischemia-reperfusion, the levels ot MDA, XOD, LDH, CK in plasma and MDA, XOD, MPO, W/D in brain tissue increased, which were maximal in LIR4h or LIR6h, then decreased. The activation of SOD in in plasma and brain tissue decreased, which was minimal in LIR4h, then increased. After LIR, the content of tNOS,iNOS in brair tissue increased, cNOS decreased. The content of NO, ET-1 in IR group in plasma and brain tissue increased, but the ratio of NO/ET-1 decreased, which was minimal in LIR4h. Conclusion: The brair injury after LIR of rats might be concerned with the increase of ROS,the decrease of antioxidant and the disturbance of the balance relationship of NO/ET-1.
