摘 要:目的通过研究Ca2+ATP酶在透明晶状体及白内障晶状体上皮细胞上的表达情况,从分子水平探讨老年性白内障的发生机制。方法利用半定量RTPCR技术测定6种Ca2+ATP酶的mRNA在透明晶状体和3种常见类型老年性白内障晶状体上皮细胞上的表达。结果6种Ca2+ATP酶在透明晶状体和白内障晶状体上皮细胞上均有表达,但表达的量不同,其中PMCA3在白内障晶状体上皮细胞上的表达明显减少,差异有显著性(P<0.01)。不同类型的老年性白内障晶状体上皮细胞上Ca2+ATP酶的表达形式亦不相同。结论PMCA3在老年性白内障的发生中可能起着关键性作用;不同形式的Ca2+ATP酶表达的减少可能导致不同类型老年性白内障的发生。Objective To explore the mechanism of cataract formation through the study of Ca^2+ -ATPase gene expression in clear and cataractous lens epithelium. Methods Half-quantity reverse transcription polymerase chain rcaction(RT-PCR) was employed to detect six different Ca^2+ -ATPases n]RXA in clear and senile cataractous lens epithelium. Results The six different Ca^2+ -ATPases were all expressed in clear and cataractous lens epithelium. But the quantities were different. The expression of the plasma membrane Ca^2+ -ATPase three (PMCA3) was decreased greatly in cataractous lens epithelium(P〈0.01). The expression of Ca^2+ -ATPases was also different in different kinds of senile cataract. Conclusion PMCA3 may play an important role in cataract fornmtion. The decrease of the expression of different Ca^2+ - ATPases may cause the formation of different kinds of senile cataract.
