姜黄素与顺铂联合对人肺腺癌A549细胞增殖抑制作用的体外研究

摘　　要：目的研究姜黄素和顺铂联合应用对人肺腺癌A549细胞增殖的影响。方法采用MTT法观察姜黄素和顺铂对人肺腺癌A549细胞的抑制作用。结果姜黄素及顺铂能抑制人肺腺癌A549细胞的增殖,并呈浓度及时间依赖性,两者作用人肺腺癌A549细胞48h的半数抑制浓度(IC50)分别为18.4μmol/L、0.966μg/ml。姜黄素10μmol/L、15μmol/L2、0μmol/L与顺铂1μg/ml、2μg/ml联合24h、48h、72h的抑制率显著高于单用顺铂、姜黄素组(P<0.05),两者呈现出相加的抗瘤效果。结论姜黄素在体外对人肺腺癌A549细胞的增殖有明显的抑制作用,能提高顺铂对人肺腺癌A549细胞的敏感性。Objective To investigate the anti-proLiferation effect of curcumin combined with cisplatin on the human lung cancer cell line A549 in vitro. Methods MTT was used to measure inhibitory effects of cureumin and cisplatin on growth of A549 cells. Results Curcumin and cisplatin inhibited the growth of the human lung cancer cell line A549 in a concentration-and time-dependent manner, their IC50 were 18.4μmol/L,0.966μg/ml respectively. Compared with either cureumin or eisplatin alone, combining cureumin at 10μmol/ L, 15μmol/L,20μmol/L with cisplatin at 1μg/ml,2μg/ml respectively increased the growth inhibition rate of A549 cells （ P 〈 0.05 ） significantly, suggesting synergistic actions of the two drugs. Conclusion Cureumin could significantly inbibit the growth of A549 ceils, which increases the sensitivity of A549 cells to cisplatin.
