摘 要:目的:分析怒、恐应激大鼠脑内c-fos、CRHmRNA表达差异机理。方法:参照Megan E与Takashi方法制作怒、恐应激动物模型,采用免疫组化法和原位杂交法,检测脑内c-fos与CRHmRNA表达,将愤怒组与恐惧组、模型组与正常组进行对照。结果:c-fos表达,模型组与正常对照组对比有显著性差异(P<0.05,P<0.01);在表达部位上,愤怒组下丘脑表达显著,而恐惧组杏仁核表达显著,两组有显著性差异(P<0.01);CRHmRNA表达主要见于下丘脑,模型组与正常对照组相比有显著性差异(P<0.05,P<0.01),愤怒组与恐惧组比较有显著性差异(P<0.01)。结论:怒、恐应激性质不同,c-fos、CRHmRNA在脑内表达部位有别。Objective: Analyse the diversity of c-fos and CRHmRNA expression in brain in rats with anger and fear stress. Methods: Animal models were established according to Megan E and Takashi's methods respectively. The expression of protein product of c-fos was examined by immunochemical technique The situs hybfidisation was used to detect the expression of CRHmRNA. Comparasion is carried out between the anger group and fear group, model group and normal group, Results : Anger and fear could significantly increase the expression of c-fos compared with normal group ( P 〈 0.05, P 〈 0.01 ) .In anger group, c-fos protein expresses especially in hypothalamus, while fear group in amygdala. The two groups vary significantly ( P 〈 0.01 ) . CRHmRNA expresses especially in hypothalamus, anger and fear could significantly increase the expression compared with the normal group ( P 〈 0.05, P 〈 0.01).The two groups vary significantly (P 〈0.01) .Conclusions: Different qualities of anger and fear stress have different expressions of c-fos and CRHmRNA in brain.
