摘  要:目的从新生大鼠大脑皮质中分离、培养并鉴定星形胶质细胞。方法分离出生1 ̄3d的大鼠大脑皮质细胞加入含有10%马血清、10%小牛血清以及10ng/mLEGF的MEM培养基培养,于第3 ̄4周将细胞固定后采用免疫细胞化学技术检测特异性星形胶质细胞抗原GFAP的表达。结果从新生大鼠大脑皮质分离的细胞生长旺盛,星形胶质细胞形态典型,并表达星形胶质细胞特异性抗原GFAP,未见其他种类细胞生长。结论笔者建立了一种简单、可靠的大鼠星形胶质细胞的体外培养方法,为应用胶质细胞饲养层与其他细胞联合培养奠定了基础。[ Objective] To isolate and identify astrocytes from cerebral cortex of neonatal rats. [Methods] The cells were isolated from cerebral cortex of neonatal rats by using MEM media added with 10% equine serum, 10% bovine serum and 10 ng/mL EGF; then the cells were fixed in the third to fourth week after culture and immunocytochemistry was used to detect specific GFAP antigen of the cells. [Result] The ceils isolated from cerebral cortex of neonatal rats grew well and expressed GFAP antigen. [Conclusion] We established a simple, reliable method of culturing and purifying astrocytes of neonatal rats and underlined the co-culturing astrocyte feeder layer with other cells culture in the future.
