细胞分裂方向的改变和Numb的表达增高在宫颈鳞癌中的作用

摘　　要：目的探讨细胞分裂方向及细胞命运决定子Numb的表达改变与宫颈鳞癌发生的关系。方法测量经病理检查确诊的59例CIⅡ-Ⅲ、21例CINⅠ及48例正常宫颈鳞状上皮的基底细胞的分裂角度,并采用免疫组织化学染色检测上述各例及50例宫颈浸润性鳞癌(ISCC)的Numb蛋白的表达结果在正常宫颈鳞状上皮中基底细胞分裂以垂直分裂为主(54.4%);在CINⅠ中,垂直分裂减少(40.3%),而水平分裂增加(32.8%);在CINⅡ-Ⅲ中,细胞分裂方向紊乱。基底细胞分裂角度在CINⅠ组和正常对照组间无显著性差异,而CINⅡ-Ⅲ组与CINⅠ组及正常对照组相比较有显著性差异。Numb的表达除在正常对照组和CINI组之间没有差异外,在各组之间有显著性差异,表现为从正常对照组到ISCC组逐渐增强。结论细胞分裂方向的改变和Numb分子的表达增高可能参与了宫颈鳞癌的发生发展过程。Objective：To investigate the relationship of the alteration of cell cleavage orientation and the expression of cell fatedeterminant Numb with the carcinogenesis of cervix squamous cell carcinomas. Methods The division orientation of the basal cells in cervix squamous epithelia were measured in pathologically documented CINⅡ-Ⅲ （n = 59）,CINI（n = 21） and normal controls （n = 48）. And Numb＇s expression was detected by immunohistochemistry in all these cases above and another 50 cases of ISCC. Resuits In normal cervix squamous epithilia, most basal cells divided vertically（54.4%）. In CINⅠ cases, the vertical divisions decreased（40.3 % ） but the horizontal divisions increased（32.8 % ）. In CINⅡ-Ⅲ cases, the division orientation of basal cells was disordered. The division orientation of the basal cells was significantly different between CINⅡ-Ⅲ group and CINⅠ group,or between CINⅡ -Ⅲ group and normal controls, but not between CINⅠ group and normal cases. The expression of Numb was significantly different between any of the two group,except between CINⅠ group and normal controls,showing an increase tendency of Numb expression from normal control to ISCC group.Conclusion The alteration of cell division orientation and the increase of Numb expression may be involved in the carcinogenesis and development of cervix squamous cell carcinomas.
