P50 auditory evoked potential in substantia nigra of normal female rats and female rat models of Parkinson disease induced by 6-hydroxydopamine

摘  要:目的:在清醒安静的自然状态下记录雌鼠黑质听觉P50诱发电位,同时观察6-羟基多巴胺对雌鼠黑质听觉P50诱发电位的影响。方法:实验于2003-01/2004-01在昆明医学院生理教研室神经电生理和帕金森病研究室进行。取成年雌性Sprague-Dawley鼠61只,在右侧黑质致密带埋置电极后随机分为3组:①模型组(n=25)雌鼠黑质致密带给予4μL的6-羟基多巴(2g/L),以0.5μL/min速度推注,注射完毕留针5min。②溶剂对照组(n=15)则向黑质致密带内注入同等剂量维生素C。③正常对照组(n=21)不给药。采用试验/条件(T/C)刺激模式,在清醒安静的自然状态下记录各组雌鼠黑质听觉P50诱发电位。结果:51只大鼠进入结果分析。①黑质内微量注射神经毒剂6-羟基多巴可成功建立帕金森病模型(皮下注射阿朴吗啡1mL/kg后,定向旋转>7r/min,持续时间>30min为造模成功);成功率60%(15/25)。②P50试验/条件(T/C)比值:正常对照组为66.68±3.71;溶剂对照组和正常对照组比较差异不显著(64.05±5.89,P>0.05),模型组显著低于溶剂对照组和正常对照组(39.90±5.41,P<0.01)。结论:①正常雌鼠黑质内存在听觉P50诱发电位。②多巴胺可能对黑质P50起着重要的调节作用。AIM: To record the P50 auditory evoked potential in substantia nigra of female rats in the quiet and awake natural states. Meanwhile, to observe the influence of 6-hydroxydopamine (6-OHDA) on the P50 auditory evoked potential in substantia nigra of female rats. METHODS: The experiment was carried out in the research rooms of neureelectrephysiology and Parkinson disease, the Department of Physiology of Kunming Medical College between January 2003 and January 2004. Sixty-one female Sprague-Dawley rats were embedded with recording electrode into right substantia nigra pars compacta, and then randomly divided into three groups: ① model group (n=25): the substantia nigra pars eompaeta of the female rats were given 4 μL 6-OHDA (2 g/L) at a speed of 0.5 μL per minute, and the needle was kept for 5 minutes after injection. ② solvent control group (n=15): Vitamin C of the same dosage was injected into substantia nigra pars compacta. ③ normal control group (n=21): no administration was given. P50 auditory evoked potentials in substantia nigra were recorded in all the three groups by means of stimulation model of testing/conditioning (T/C). RESULTS: Totally 51 rats were involved idn the analysis of results. ① 6-OHDA micro-injection in substantia nigra pars compacta can establish Parkinson disease model successfully (After subcutaneous injection of 1 mL/kg apomorphine, the oriented rotation 〉 7 round per mintes and lasted for 〉 30 minutes were taken as successful model establishment), and the successful rate was 60% (25/50). ② Testing/conditioning (T/C) ratio of P50 in the normal control group was 66.68 ±3.71, which had no significant diffenence from that in the solvant control group (64.05±5.89, P 〉 0.05), and it was significantly lower in the model group than in the solvant control group and normal control group (39.90±5.41, P 〈 0.01).
