摘 要:目的测定无梗五加果中总香豆素及6,7-二甲氧基香豆素的含量。方法以东莨菪内酯为对照品,采用分光光度法,于348nm处测定总香豆素含量。采用HPLC法测定6,7-二甲氧基香豆素含量。色谱柱为Hypersil BDS C18(4.6mm×250mm,5 μm),流动相为甲醇-磷酸水溶液(pH3.0)-乙腈-四氢呋喃(体积比为15:77:6:2),检测波长为340nm。结果总香豆素质量浓度在3.44~12.9mg·L^-1(r=0.9992)内、6,7-二甲氧基香豆素质量浓度在1~40mg·L^-1(r=0.9999)内线性关系良好;平均回收率分别为100.6%、97.7%;总香豆素和6,7-二甲氧基香豆素的质量百分含量分别为0.12%、0.0037%。结论方法准确、可靠,可为无梗五加果的质量控制提供参考依据。Objective To determine the contents of total coumarin and 6, 7-dimethoxycoumarin in fruit of Acanthopanax sessiliflorum (Rupr. et Maxim. )Seem. Methods The content of total coumarin was determined with ultraviolet spectrophotometry at 348 nm using scopoletine as reference. The separation and quantitation of 6, 7-dimethoxycoumarin was performed on a Hypersil BDS C18(4.6 mm × 250 mm, 5μm)with mobile phase consisted of methanol-H3PO4 (pH3.0)-acetonitrile-tetrahydrofuran ( V : V : V: V = 15 : 77 : 6 : 2) and detection at 340 nm. The flow rate was 0.8 mL·min^-1. Results The standard curve was linear in the range of 3.44- 12.9 mg·L^-1 for total coumarin(r = 0. 999 2)and 1 -40 mg·L^-1 for 6, 7-dimethoxycoumarin (r = 0. 999 9). The average,recoveries of total coumarin and 6, 7-dimethoxycoumarin were 100.6 % and 97.7%, respectively. Conclusions The methods are simple, accurate with good reproducibility and can be used to assess the quality of fruit of Acanthopanax sessiliflorum (Rupr. et Maxim.)Seem.
