脐血树突状细胞介导的食管癌疫苗抗肿瘤免疫预防实验研究

摘　　要：目的用杂交瘤技术制备脐血树突状细胞(DC)和食管癌细胞的融合疫苗,观察其在体内诱导抗肿瘤免疫反应。方法分离脐血CD34+干细胞并诱导扩增为成熟DC,并与EC109细胞经聚乙二醇(PEG)法融合;免疫磁珠法筛选EC109-DC;重建SCID小鼠免疫功能;体内保护抗肿瘤免疫保护实验。结果①融合疫苗可体外生长,形状以不规则形为主,核仁大、细胞边缘可见突起;②重建SCID小鼠免疫功能实验中PBL组均检测到人IgG的存在,与PBS组相比较有显著意义(P<0.05);③在免疫保护中,经融合瘤苗免疫的小鼠,肿瘤潜伏期较对照组明显延长(P<0.05),肿瘤大小和肿瘤重量均明显小于对照组(P<0.05)。结论①重建SCID小鼠免疫功能实验中PBL组均检测到人IgG的存在,提示免疫重建成功;②融合瘤苗对EC109细胞的攻击有明显的抵抗作用。Objective To develop fusion vaccine of esophageal carcinoma cell and DC , and to study its protective effect against EC109 cell. Methods Fusion vaccine was produced by traditional PEG fusing, cytokine inducing, CD34^＋ magnetic microbead marker sorting and MACS. Immune responses of SCID was estabished by human PBL. For studying the protective effect of the fusion vaccine against tumor EC109, 20 SCID mices were divided into P, E, D and El） subgroups, immunized by PBS, inactivated EC109, DC and the fusion vaccine respectively, and were attacked by EC109 cell. The tumor size, weight, mouse survival period and tumor latent period were recorded and analyzed. Pathology was also applied to study the protective effect of the fusion vaccine against EC109 cell. Results （1）The EC109 - DC could proliferate slowly in vitro. （2）In the experiment of immune reconstrusction, the existence of human IgG was detectedin the PBL group, significaatly different from PBS group（ P 〈 0.05）. （3）In the immune group, latent period was longer in ED subgroup than others. Tumor size and tumor weight were also smaller in ED subgroup. Conclusion （1）The existence of human IgG in the PBL group suggests that immunity is successfully reconstructed. （2）The fusion vaccine has protective effect against tumor EC109.
