摘 要:目的筛选和克隆与葡萄胎发病相关的新基因。方法采用抑制性消减杂交方法,建立葡萄胎组织与正常早孕绒毛组织间的差示cDNA文库,从中筛选出新基因序列,并克隆其全长。结果从差示cDNA文库中共筛选出28个克隆,测序结果提示含10种基因序列,其中3个基因片段被确认为功能未明的新基因,并获得其中一条基因的全长序列(按克隆序列号命名为F10)。结论F10等新基因可能与葡萄胎的病理发生发展密切相关。Objective To screen and clone new genes that are associated with the pathogenesis of hydatidiform mole. Method Suppressive subtracfive hybridization method was applied. Subtractive cDNA library was made between hydatidiform mole and normal early - pregnancy villous tissues. New genes were screened and full - length sequence was cloned. Results Totally 28 clones were selected from the cDNA library, 10 gene sequences were identified. Among these clones, 3 genes were found to be new genes. And the full length sequence of one gene F10 were cloned. Conclusion F10 gene might be closely associated with the pathogenesis of hydatidiform mole.
