老年期抑郁症和轻度阿尔茨海默病的血脂和载脂蛋白E基因多态性研究

摘　　要：目的探讨血脂及载脂蛋白E(ApoE)基因多态性与老年期抑郁症和轻度阿尔茨海默病(AD)的关系。方法采用PCR-RFLP技术测定60例老年期抑郁症、30例轻度AD患者和60例正常人的ApoE基因型,采用酶法检测3组受检者的血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL-C)和低密度脂蛋白(LDL-C)。结果老年期抑郁症和轻度AD患者的TC和TG均显著高于正常对照者,而LDL-C均显著低于正常对照者(P<0.05或P<0.01)。老年期抑郁症患者与正常对照者ApoE的基因型和等位基因频率分布间差别无显著性意义(P>0.05);轻度AD患者的ApoEε4等位基因频率显著高于正常对照者(P=0.011)。结论老年期抑郁症和轻度AD患者均存在脂质代谢的异常,ApoE基因型可能在老年期抑郁症的发病中不起重要作用。Objective To investigate the relationship of serum lipid concentrations and apolipoprotein E genotype among patients with senile depression and alzheimer disease. Methods Apolipoprotein E genotype was determined in 60 patients with senile depression, 30 patients with alzheimer disease and 60 healthy controls by using PCR - RFLP assay. Serum concentrations of total cholesterol （ TC）, triglycerides （ TG）, high - density lipoprotein cholesterol （ HDL - C） and low - density lipoprotein cholesterol （ LDL - C） were examined by enzymatic methods. Results Patients with senile depression and alzheimer disease were significantly higher in TC and TG than healthy controls, significantly lower in LDL - C than healthy controls （ P 〈 0. 05 or P 〈 0. 01 ）. There were no significant differences in ApoE polymorphism genotypes and alleles among patients with senile depression, alzheimer disease and healthy controls （ P 〉 0. 05 ）. Compared with healthy controls, there were significant increase of ApoEε4 in patients with alzheimer disease （ P = 0. 011 ）. Conclusion Patients with senile depression and alzheimer disease have severe abnormality in metabolism of lipid. These results show the ApoE gene may be not important in the pathogenesis of senile depression and alzheimer disease.
