摘 要:目的观察斑蝥灸对肿瘤抑制率和肿瘤转移抑制基因nm23-1的m RNA表达的影响。方法实验于2004-05/08在针灸学院实验室和广安门医院分子生物实验室进行。实验动物C57BL/6J近交系小鼠50只,雌雄各半。随机分成5组,每组10只。正常对照组不做造模及治疗处理;肺癌模型组接种肺癌瘤株,不做治疗处理;单纯化疗组;单纯斑蝥灸组;斑蝥灸+化疗组。后4组接种Lewis肺癌瘤株,建立Lewis肺癌模型。用实时荧光定量反转录聚合酶链反应技术检测肺癌小鼠nm23-1基因的m RNA表达。结果各组小鼠nm23-1基因的m RNA表达量和nm23-1与β-actin基因的m RNA表达5组动物45只进行结果分析。①斑蝥灸组显著提高Lewis肺癌小鼠nm23-1基因的m RNA表达,与各自β-actin基因的m RNA表达比值的均值为5.68。②单纯化疗组和斑蝥灸配合化疗组与对照组比较,差异无显著性意义,但斑蝥灸配合化疗组明显优于单纯化疗组,其均值分别为4.35和1.8。③斑蝥灸配合化疗组对Lewis肺癌的抑制率是40.55%,优于单纯化疗组的31.09%。结论斑蝥灸配合化疗对Lewis肺癌的生长有明显的抑制作用,其作用优于单纯化疗组,说明斑蝥灸可以增强化疗药物的抗癌作用。其作用可能与促进nm23-1基因的m RNA表达有关。AIM: To observe the effects of moxibustion of cantharidin on the expression of mRNA of nm23-1 gene. METHODS: The experiment was conducted at the laboratory of Acupuncture College and biochemical laboratory of Guanganmen Hospital from May to August 2004. Experimental animals: Fifty C57BL/6J inbred line male and female (half and half) mice were selected, and assigned randomly into 5 groups with 10 rats in each group. Normal control group: The rats were not treated with therapy or building models; Lung cancer model group: The rats were inoculated with tumor of lung cancer without therapy; Pure chemiotherapy group; Pure cantharidin group; Cantharidin + chemiotherapy group. The rats in the later 4 groups were inoculated with Lewis lung cancer tumor to build Lewis lung cancer model. The expression of mRNA nm23-1 gene in lung cancer mice was detected with real-time fluorescent quantitative reverse transcription polymerase chain reaction (RT-PCR) method. RESULTS: The expression of mRNA of nm23-1 gene in mice of every group and nm23-1 with expression of mRNA of β-actin gene in 45 animals of 5 groups were involved in the result analysis. (1) The expression of mRNA of nm23-1 gene in Lewis lung cancer mice enhanced significantly in the cantharidin group, and the mean value of ratio with the expression of mRNA of every β-actin was 5.68. (2) The difference had insignificant meaning as compared between pure chemiotherapy group and cantharidin + chemiotherapy group with control group. But it was better in the cantharidin + chemiotherapy group than that in the pure chemiotherapy group, and its mean value was 4.35 and 1.8, respectively. (3) The inhibitive rate of Lewis lung cancer in the cantharidin + chemiotherapy group was 40.55%, which was better than 31.09% in the pure chemiotherapy group. CONCLUSION: The cantharidin matching with chemiotherapy has obviously inhibitive effects on the growth of Lewis lung cancer, which is better than that in the pure chemiotherapy group. It is indicat
