胃癌组织中血管内皮生长因子和基质金属蛋白酶-9的表达及相关性研究

摘　　要：目的探讨胃癌中血管内皮生长因子(VEGF)和基质金属蛋白酶-9(MMP-9)的表达与胃癌生物学行为的关系。方法应用免疫组织化学SP法检测70例胃癌组织及38例远癌胃黏膜中VEGF与MMP-9的表达。结果VEGF在胃癌中的表达(61.4%)显著高于远癌胃黏膜的表达(31.6%)(P<0.01),且与淋巴结转移呈正相关(P<0.01),而与年龄、性别、组织分化程度及肿块浸润深度无关(均P>0.05)。MMP-9在胃癌中的表达(68.6%)显著高于远癌胃黏膜的表达(39.5%)(P<0.01),且与淋巴结转移呈正相关(P<0.05),而与年龄、性别、组织分化程度及癌组织浸润深度无关(均P>0.05)。结论MMP-9、VEGF在肿瘤浸润、转移中起重要作用;其共位表达有助于判断胃癌预后。Objective To identify the expression of vascular endothelial growth factor（VEGF） and matrix metalloproteinase 9（MMP-9） in gastric carcinoma and its relationship with the biological behavior of gastric carcinoma. Methods The expression of VEGF and MMP-9 in 70 cases of gastric carcinoma tissue and 38 cases of gastric mucosa distant from carcinoma were detected by streptavidin peroxidase immunohistochemistry. Resulls The expreesion of VEGF in gastric carcinoma（61.4 % ） was sigificantly higher than that in gastric mucosa distant from carcinoma（31.6 % ）（P〈 0.01 ）. The expression of VEGF was positively correlated with lymph node metastasis（ P 〈 0,01）, but was not related to age,sex of the patients, and the degree of histological differentition or depth of tumor infiltration（ P 〉 0,05）. The expreesion of MMP-9 in gastric carcinoma（68.6 % ） was significantly higher than that in gastric mucosa distant from carcinoma（39.5 % ）（ P 〈 0.01 ） and was positively correlated with lymph node metastasis（P 〈 0.05）, but not related to age, sex of the patients, and the degree of histological differentiation or depth of tumor infiltration（P 〉0,05）, Conclusions MMP-9 mRNA and VEGF expression in gastric carcinoma appears to be well-characterlzed indicators of prognosis and metastasis.
