摘　　要：目的观察葡萄糖神经酰胺合酶(glucosylceram ide synthase,GCS)及其基因在不同的良恶性乳房肿瘤中的表达情况,以探讨GCS与乳房肿瘤的关系。方法采用免疫荧光抗体染色技术检测临床良恶性乳房肿瘤组织切片中的GCS含量;并通过RT-PCR的方法分析不同乳房肿瘤组织中GCS基因的表达情况。结果恶性肿瘤组织切片中GCS的表达明显强于良性肿瘤组织:且以β-actin为内标,恶性肿瘤组织GCS基因的相对表达量与良性组织相比亦存在显著性差异(P<0.05)。结论乳房癌组织中GCS的含量显著增高,其可能与恶性肿瘤细胞的无限制生长及产生耐药性相关。Objective： To investigate the expression of glucosylceramide synthase in breast tumour and relationship each other. Methods ： Using immunofluorescence assay the expression of GCS in clinical samples, which included breast cancer and fibroadenoma. was detected. GCS mRNA was also analyzed by RT - PCR. Results ： Compared with breast fibroadenoma, the expression of GCS and relative quantities of GCS mRNA in breast cancer exhibited significantly increased. Conclusion ： The high level of GCS in breast cancer is possible correlated with developing and multidrug resistance of cancer cells.
