摘  要:目的:探讨药物性肝损伤的发生机理及保肝解毒颗粒的作用机制。方法:60只小鼠随机分为模型组、保肝解毒颗粒大、中、小剂量组、甘利欣对照组和空白对照组,分别以相应剂量药物灌胃5天,然后以四环素灌胃造模,检测小鼠血清ALT、AST和IL-18水平及肝细胞凋亡情况。结果:模型组小鼠血清IL-18水平明显升高,肝细胞凋亡程度严重;保肝解毒颗粒可有效降低血清ALT、AST和IL-18水平,并使肝细胞凋亡程度明显减轻。结论:四环素所致的急性肝损伤与IL-18和肝细胞凋亡密切相关;保肝解毒颗粒可通过降低血清IL-18水平、抑制肝细胞凋亡等起到防治肝脏损害的作用。Objective: To explore the mechanisms of drug-induced liver injury and the acting mechanisms of Baogan Jiedu Granule. Method:60 mice were randomly divided into Tetracycline model group, big-dosage, medium- dosage and small-dosage group of Baogan Jiedu Granule, Ganlixin group and control group. Each was respectively perfused with corresponding drug for 5 days and Tetracycline was perfused once to make model, The levels of ALT,AST and IL- 18 in the serum were respectively measured and liver cell apoptosis was observed. Result: IL- 18 in the serum of model group rised obviously and the mice appeared to be severe apoptosis of the liver cells. Baogan Jiedu Granule could reduce the level of ALT,AST and IL- 18 obviously and showed great effect on restraining cell apoptosis. Conclusion;Acute liver injury induced by Tetracycline has close relation to IL- 18 and cell apoptosis. The mechanism of Baogan Jiedu Granule on preventing and curing acute liver injury is mainly relative to reducing the level of IL- 18 and inhibiting apoptosis of the liver cells.
