MMP-2表达与肝细胞癌侵袭、转移的关系研究

摘　　要：目的:探讨人肝细胞癌中基质金属蛋白酶-2(m atrix m eta lloprote inase-2,MM P-2)的表达与肝癌侵袭、转移的关系。方法:选择52例有完整随访资料的原发性肝癌及相应癌旁组织标本、20例正常肝组织标本,用RT-PCR方法检测癌、癌旁组织及正常组织中MM P-2 mRNA的表达。结果:①MM P-2 mRNA在肝癌中的表达高于癌旁组织和正常组织,差异有统计学意义(P<0.05),癌旁组织和正常组织中MM P-2 mRNA的表达差异无统计学意义(P>0.05);②癌组织中MM P-2 mRNA表达与术后复发时间呈负相关(r2=0.868,P<0.05),与病理学分级呈正相关(r2=0.786,P<0.05);③癌组织中MM P-2 mRNA的表达与肝外转移呈正相关(r2=0.812,P<0.05);④癌旁组织中MM P-2 mRNA的表达与术后复发时间呈负相关(r2=0.854,P<0.05)。结论:MM P-2 mRNA与肿瘤的分化程度,侵袭、转移能力,复发倾向相关,在临床中可望作为肿瘤分化、复发、转移的评估指标之一。Objective：To investigate the association between matrix metalloproteinase （MMP-2） expression of hepatocellular carcinoma （HCC） and its invasion and metastasis. Methods.RT-PCR analysis technology was used to detect MMP-2 mRNA expression in 56 paired HCC and non-HCC liver tissue samples and 20 normal liver tissue samples. Result ：①The level of MMP-2 mRNA expression in HCC was higher than that in adjacent non-HCC liver tissue and in normal liver tissue and the difference was statistically significant （P （0.05）; the difference of MMP-2 mRNA expression between adjacent non-HCC liver tissue and normal liver tissue had no statistically significance （ P 〉0. 05） ;②MMP-2 mRNA expression in HCC had a inverse correlation with recurrence after surgical treatment （r^2= 0. 868, P 〈0.05） and a positive correlation with pathological grade （r^2= 0. 786, P 〈0. 05）;③There was an positive correlation between MMP-2 mRNA expression in HCC and extrahepatic metastasis （r^2= 0. 812, P 〈0.05）;④Significant inverse correlations were found between MMP-2 mRNA expression in adjacent non HCC liver tissue and extrahepatic metastasis （r^2Z=0. 854, P〈0.05）. Conclusion：MMP-2 mRNA had an association with degree of tumor differentiation ,capability of invasion and metastasis and tendency of recurrence. It is hopeful that MMP-2 mRNA will be one of the indexes for differentiation, recurrence and metastasis.
