摘  要:目的辐射对肝癌细胞多药耐药性的影响。方法取肝癌细胞株HepG2及HepG2/ADM分别进行X线照射(10Gy),于照射前后分别采用流式细胞仪检测各组细胞表面Pgp变化,采用RT-PCR法检测MDR1表达变化。结果照射前后HepG2细胞表面P-gp阳性表达率分别为:(11.3±1.2)%(、27.4±3.9)%;HepG2/ADM组为:(45.5±5.1)%,(48.1±6.1)%。HepG2组P-gp在细胞膜上的表达较照射前后差异有显著性意义(P<0.01)。结论辐射可诱导肝癌细胞HepG2 mdr1基因表达增强,细胞膜表面Pgp表达增加。对HepG2/ADM耐药性影响较小。Objective to study the effect on expression rate of muhidrug resistance gene in hepatocular carcinoma cells after irradiation exposure.Methods HepG2 and HepG2/ADM cells were irradiated respectively ( 10 Gy)and the change of Pgp and MDR1 mRNA were detected by flow Cytometry and RT-PCR before and after irradiation respectively.Results The positive rate of Pgp before and after irradiation was( 11.3 ± 1.2) %, (27.4 ± 3.9) % for HepG2 and (45.5 ± 5.1 ) %, (48.1 ± 6.1 )% for HepG2/ADM respectively. There was a significance difference in the HepG2 groups before and after irradiation. Conclusion irradiation can induce overexpression of mdrl gone and Pgp in HepG2 cell line after irradiation exposure, while in HepG2/ADM cell line, the effect has no significance.
