胃癌组织中PTEN、PI3K的蛋白表达及意义

摘　　要：目的：主要探讨PTEN，PI3K在胃癌中的蛋白表达及其与胃癌临床病理特征的关系，以及二者表达之间的相关性。为胃癌的临床早期诊断和基因治疗提供理论依据。方法：采用免疫组织化学Elivision法检测10例正常胃粘膜组织和68例胃癌组织中PTEN、PI3K的蛋白表达。结果：1.PTEN的蛋白表达：胃癌组织中PTEN蛋白的阳性表达率为48．53％（33／68）。早期胃癌、中晚期胃癌和正常胃粘膜组织中的PTEN的阳性表达率分别为64．29％、42．87％和100．00％。早期胃癌、中晚期胃癌中PTEN蛋白的阳性表达率均较正常胃粘膜组织低，差异有显著性（p〈0．05）。PTEN蛋白的阳性表达与胃癌癌细胞的分化程度（p=0．015）及淋巴结转移相关（p=0．014）；2．PI3K的蛋白表达：胃癌组织中PI3K蛋白总的阳性表达率为85．29％（58／68）。早期胃癌、中晚期胃癌中PI3K蛋白的阳性表达率均较正常胃粘膜组织高，差异有显著性（p〈0．05）；3．胃癌组织中PTEN、PI3K蛋白表达之间的关系：本研究的68例胃癌组织中PI3K蛋白阳性表达的58例中，34例PTEN蛋白却呈阴性表达；10例PI3K蛋白阴性表达的胃癌组织．其中9例PTEN蛋白呈阳性表达．二者旱屁著负相关（p=0．006）。结论：1．PTEN蛋白的缺失与胃癌发生、发展有关。且与弥漫型胃癌发生发展的关系更加密切。PTEN蛋白可作为评价胃癌生物学行为的重要参考指标；2．PI3K蛋白的表达与胃癌的形成有关．且与胃癌癌细胞的转化过程以及肿瘤细胞的侵袭转移有关；3．PI3K、PTEN在胃癌的发生发展中的作用可能互相拮抗。Objective： This study was conducted to investigate the expression of FFEN ,PI3K （the phosphoinositide-3 kinase） proteins in gastric carcinoma and their relationship between their expression and clinicopathological parameter,and correlation among PTEN and PI3K protein expression each other,and it can afford theory bases to clioical early diagnosis and gene therapy of gastric carcinoma, Methods：Elivision immunohistochemical method was used to detect expressions of PTEN,PI3K proteins in 10 normal gastric mucosa epithelium and 68 gastric carcinoma tissues.Results：1.Expression of PTEN protein：PTEN protein positive rates was 48.53%（33/68）in gastric carcinoma tissues. In early gastric carcinoma, moderate and late gastric carcinoma, normal gastric mucosa epithelium,positive rates of PTEN protein was 64.29%,42.87% and 100.00% respectively. Early gastric carcinoma,moderate and late gastric carcinoma compared with normal gastric mucosa epithelium, there were significant differences of PTEN protein expression （p〈 0.05）.No significant difference of PTEN protein expression was observed between early gastric carcinoma and moderate and late gastric carcinoma. There were significant correlation between PTEN protein expression and lymph node metastasis（p=0.014）,cell differentiation degree （p=0.015）. 2. Expression of PI3K protein： PI3K protein positive rates was 85.29%（58/68）in gastric carcinoma tissues. Early gastric carcinoma, moderate and late gastric carcinoma compared with normal gastric mucosa epithelium, there were significant increase of PI3K protein expression（p〈0.05）. 3. Correlation among PTEN and PI3K protein expression each other： In 58 gastric carcinoma with PI3K protein positive expression,bnt negative expression of PTEN protein were detected in 34 of them;in 10 gastric carcinoma with PI3K protein negative expression,but positive expression of PTEN protein were detected in 9 of them,a highly inverse correlation was present between PI3K and PTEN protein expressions （p=0.006
