摘  要:目的研究人干扰素-α对人肝癌细胞系BEL-7402增殖及转移的抑制作用,探讨其抗肿瘤作用的机制。方法常规培养的人肝癌细胞株BEL-7402细胞加入人干扰素-α共同孵育,作用24h后,以MTT法检测干扰素-α对肿瘤细胞增殖的影响;Boyden小室法检测肝癌细胞侵袭能力的变化。体内实验中以BEL-7402细胞106个皮下接种于BALB/C裸鼠,肉眼可见成瘤后以干扰素-α隔天瘤旁皮下注射,观察小鼠的一般状态并测量肿瘤体积的变化;小鼠处死后,HE染色检测肿瘤细胞向附近淋巴结及其它脏器的转移情况,免疫组化法检测肿瘤细胞增殖核抗原的表达;Tunel法检测肿瘤细胞的凋亡。结果人干扰素-α可以明显抑制体外BEL-7402细胞的增殖(抑制率为20.2%);干扰素-α作用后在Boyden小室穿膜的肿瘤细胞数明显减少;干扰素-α治疗组荷瘤小鼠肿瘤体积明显小于对照组;所有小鼠都有近侧腋窝淋巴结的肿瘤转移,未见有其它脏器的转移;干扰素-α组裸鼠生长肿瘤细胞增殖核抗原的表达明显下降,凋亡明显增加。结论干扰素-α可以直接抑制体内、外肝癌细胞株BEL-7402的增殖,对BEL-7402细胞的侵袭有明显的抑制作用;可以抑制裸鼠移植肿瘤细胞增殖核抗原的表达并诱导细胞凋亡;但对BEL-7402细胞局部淋巴结转移无抑制作用。[Objeαive] To investigate the effeα and the relevant mechanisms of interferon(IFN)-α on proliferation and invasion of human hepatocarcinoma cell line BEL-7402 cells in vitro and in vivo. [Mαhods] BEL-7402 cells were incubated with IFN-α for 24 hours, MTT method was used to measure the cells proliferation. Boyden chambers were used to measure the capability of invasion. For in vivo studies, nude mice were implanted s.c. with BEL-7402 cells. After bearing tumor, nude mice were injeαed para-tumor s.c. with IFN-α once every interval day. Tumor dimension was dαermined by caliper. After animals were killed, tumor tissues and different organs (lung, liver, spleen and axillary nodes) were immediately removed and fixed in formaldehyde. Mαastasis was dαeαed by HE staining. Immunohistochemical technique was used to analyze PCNA expression. The apoptosis of tumor cells were observed by Tunel techniques. [Results] IFN-α can significantly inhibit the proliferation of BEL-7402 cells(the inhibiting rate was 20.2%). The number of invasive tumor cells in IFN-a group was significantly reduced than that of the controls in vitro. Tumor dinrension was significantly reduced in IFN-α groups. HE staining showed that BEL- 7402 cells can migrated to axillary nodes but not to other organs in all mice. PCNA expression of tumor tissues was much lower and the apoptosis of tumor cells were increasing significandy than those of controls. [Conclusion] IFN- α can inhibit the proliferation of BEL-7402 cells in vitro and in vivo. It inhibited the invasion in vitro but didn' t show any inhibiting effeα on mαastasis of BEL-7402 cells in vivo. IFN-α can reduce the expression of PCNA and induce the apoptosis of tumor cells in nude mice.
