摘  要:目的探讨LRIG1在人星形细胞瘤中的表达与意义。方法设计、制作LRIG1mRNA寡核苷酸探针,用原位杂交检测LRIG1mRNA在16例不同级别人星形细胞瘤组织及11例原代培养的人脑星形细胞瘤细胞中的表达,同时用免疫组化检测增殖细胞核抗原(PCNA)的表达,分析肿瘤LRIG1表达和PCNA表达之间的关系。结果人星形细胞瘤组织和培养的星形细胞瘤细胞中均有一定程度的LRIG1表达,其表达的程度与PCNA表达呈显著负相关(P<0.05)。结论LRIG1蛋白具有抑制星形细胞瘤增殖的作用,可能参与了肿瘤的发生和发展过程。Objective To investigate the expression of the LRIG1 gene in human astrocytomas and its significance. Methods A triphasic oligonucleotide probe was designed. The expression level of the LRIG1 gene was determined by in situ hybridization in 16 cases of astrocytomas and 11 cases of primary culture astrocytomas cells. The expression of proliferating cells nuclear antigen (PCNA) was detected by immunohistochemistry in these tissues. The relationship between both the LRIGI and PCNA was analyzed. Results There was LRIG1 gene expression in both the human astrocytomas and cultured astrocytoma cells. The expression of LRIG1 gene was negatively correlated to the expression of PCNA in both the astrocytomas and cultured astrocytomas ceils (P〈0.05). Conclusion LRIG1 may play a role in reducing the tumor's proliferation and take part in the pathogenesis and development of the astrocytomas.
