p15在食管癌中的表达及生物学意义

摘　　要：目的探讨p15 mRNA及其蛋白在食管癌中的表达与食管癌发生的关系。方法采用原位杂交(insuit hybridyzation)技术和链霉卵白素-过氧化物酶(streptavidin-horseradish peroxidase,SP)免疫组织化学染色法检测48例食管鳞状细胞癌组织,18例癌旁不典型增生组织和10例切缘正常组织中p15 mRNA及其蛋白的表达水平。结果p15 mRNA及其蛋白在食管癌组织中的阳性表达率分别为56.3%和47.9%,二者表达呈明显正相关(χ2=22,P<0.05),均明显低于在正常组织和不典型增生组织中的表达(P<0.05);且p15 mRNA及其蛋白的表达与肿瘤的分化程度相关(P<0.05)。结论p15低表达可能与食管癌的发生有关,p15表达对食管癌恶性程度的评估有一定的参考价值。Objective To investigate p15 mRNA and p15 protein expression in the esophagus cancer and the putative roles in carcinogenesis of esophagus cancer. Methods The expression of p15 mRNA and p15 protein was examined respectively by in suit hybridyzation （ISH） and streptavidin-horseradish peroxidase（SP） immunohistochemistry（IHC） method in 48 cases of esophageal carcinoma, 18 cases of esophageal dysplasia and 10 cases of incisal margin normal tissues. Results The positive expression rates of p15 mRNA and p15 protein in the esophagus cancer were 56.3 % and 47.9 %, which were respectively lower than those in incisal margin normal tissues and adjacent dysplasia tissues（ P 〈0.05）. There was positive correlation between the expression of p15 mRNA and p15 protein in the esophageal carcinoma tissues（X^2=22, P 〈0.05）. The expression of p15 had corre Conclusion The low expression of p15 might invo ation with histological difference（ P〈0.05）. ve in esophageal carcinogenesis. There is some reference value of p15 expression to the malignancy degree of esophageal cancer.
