摘 要:目的研究树突状细胞(DC)瘤苗的抗肿瘤免疫保护治疗作用,进一步探索DC为基础的肿瘤免疫治疗的新途径。方法以腺病毒为载体转染IL-18和/或gp100-DC,制备DC瘤苗,以ELISA、RT-PCR、FACS及混合淋巴细胞反应(MLR)检测所制备的DC瘤苗的效果,以黑色素肿瘤动物模型鉴定DC瘤苗的抗肿瘤免疫保护作用。结果本研究成功制备了DC瘤苗,IL-18/gp100-DC的抗肿瘤免疫保护作用明显强于IL-18-DC或gp100-DC瘤苗。结论Th1型细胞因子IL-18和gp100共转染能显著增强DC的抗肿瘤作用。Objective To explore the anti - tumor effect of dendritic cells (DC) vaccine and new immunotherapy method. Methods Cytokine interleukin - 18 (IL- 18) an melanoma- associated antigen gpl00 were transfectod into bone marrow- derived DC (IL- 18/gp100- DC), which were used as vaccine to induce the protective in a B16 melanoma model. And the effect of DC vaccine were detected through ELISA, RT - PCR, FACS and MLR. Results DC vaccines were successful constructed. The effect of anti - tumor immunity in IL - 18/gp100 - DC was significant higher than that of IL - 18 - DC or gp100 - DC. Conclusion The effect of anati - tumor was significanly enhanced with DC vaccine co - expressing Th1 cytokine IL- 18 and gp100.
