摘 要:目的全反式维甲酸(all trans retinoic acid,ATRA)对多种肿瘤细胞具有明显的诱导分化作用,对人子宫内膜癌细胞亦如此,本文通过观察ATRA对子宫内膜腺癌细胞株(RL95-2)中基质金属蛋白酶-9(matrix metalloproteinase 9,MMP-9)的作用,进一步探讨ATRA对肿瘤细胞浸润性的影响。方法体外培养的子宫内膜癌细胞分为两组,一组给予ATRA处理,另一组设为对照组,用免疫组织化学方法检测两组RL95-2细胞中MMP-9的表达。结果经ATRA处理后的子宫内膜癌细胞中MMP-9呈阴性表达,未用药组MMP-9呈阳性表达,定位于细胞浆。结论ATRA对体外培养的RL95-2细胞中MMP-9的表达具有抑制作用,可能降低子宫内膜癌细胞的浸润性。Objective To evaluate the action of all trans retinoic acid(ATRA) on matrix metalloproteinase 9 (MMP 9) expresion in RL95-2 so as to observe the role of ATRA in affecting invasion of RL95-2. Mehtods RL95-2 cultured in vitro was divided into two groups, treatment group (treated with ATRA) and control group, in which the expression of MMP-9 in RL95-2 was evaluated by immunohistochemical method at specific time point after treatment with ATRA. Results MMP-9 could be found in cytoplasm in control group rather than in treatment group. Conclusion ATRA has inhibitive effect on MMP9 expression in RL95-2 cultured in vitro and may lead to a reduction of invasive capacity of RL9512.
