摘 要:目的观察姜黄素对肺腺癌细胞A549生长的抑制作用,并探索其机制。方法四氮唑蓝法测定姜黄素作用于肺腺癌细胞A549的吸光值,计算细胞抑制率;流式细胞仪测定40μmol.L-1姜黄素或空白对照培养液的肺腺癌细胞凋亡率与凋亡相关蛋白Caspase-9表达;比色法测定凋亡相关蛋白Caspase-3,Caspase-8表达;观察加用Caspase-9抑制剂后Caspase-3的表达。结果姜黄素对肺腺癌A549细胞有抑制作用,存在剂量依赖(r=0.619;P<0.01)与时间依赖(r=0.618;P<0.01);姜黄素对肺腺癌A549的凋亡率(30.25%)明显高于对照组(1.32%)(P<0.001);姜黄素组肺腺癌细胞A549的凋亡相关因子Caspase-3、Caspase-8、Caspase-9表达明显高于对照组(P<0.001);姜黄素组加用Caspase-9抑制剂后Caspase-3表达明显下降,仍高于对照组(P<0.001)。结论姜黄素诱导肺腺癌A549细胞凋亡可能与激活Caspase-8、Caspase-9、Caspase-3有关。AIM To explore the effect of cureumin on inhibiting the apoptosis of the lung adenocarcinoma A549 cells and related mechanism. METHODS The inhibiting ratio of curcumin in different concentrations was determined by MTr assay at different times;The apoptosis rates of A549 cells cultured with 40 μmol·L^-1 curcumin were calculated by Flowcytometry. The Caspase-9 expression of each group was determined by Flowcytometry. The Caspase-8 and Caspase-3 expression of each group were determined by colorimetry set. The Caspase-3 expressions of curcumin groups added Caspase-9 inhibitor were determined in the same way. Statistical method: Z2 test or t test with SPSS10.0 software. RESULTS The inhibition of curcumin on lung adenocarcinoma A549 cells was shown in the dose-dependent and time-dependent manner( P 〈 0.01 ). The apoptosis rate of A549 cells induced by curcumin (30.25 % )was remarkably higher than the control(1.32%)( P 〈 0.001 ). The expressions of Caspase-3,-8,-9 induced by cureumin were obvious compared with the control ( P 〈 0.001 ). After using Caspase-9 inhibitor , the expression of Caspase-3 decreased remarkably ( P 〈 0. 001 ), but it was more than control group( P 〈 0. 001 ). CONCLUSION Curcumin can activate the expression of Caspase-8, Caspase-9 and Caspase-3 and induce the apoptosis of lung adenocarcinoma A549 cells through mitochondrion-depended and mitochondrion-undepended pathway.
