摘 要:目的探讨大鼠颞叶癫痫发作后不同时间学习、记忆障碍程度与海马病理变化的相关性。方法采用立体定向技术在大鼠右侧海马注射红藻氨酸诱发颞叶癫痫发作,观察其行为学表现、脑电图变化,M orris水迷宫评价不同时间段学习、记忆障碍程度,以及海马、皮质病理变化。结果注射红藻氨酸大鼠出现颞叶癫痫发作,随发作时间延长出现不同程度学习、记忆障碍,定位航行实验示逃避潜伏期延长,空间搜索实验示原平台象限内游泳时间百分比下降,与对照组比较均有显著性差异(P<0.05),发作后2个月达到高峰。右侧海马锥体细胞逐渐出现变性、坏死,由CA3区向CA4、CA2、CA1区扩展,2个月达到高峰;对侧海马锥体细胞也出现变性、坏死,但程度明显低于注射侧。结论大鼠颞叶癫痫发作后,其学习、记忆障碍程度与海马变性、坏死程度具有一定相关性。Objective: To explore the correlation between learning and memory impairment and pathological change of hippocampus in different time after temporal lobe epilepsy in rats. Methods: Kainic acid was injected into right hippocampus to induce temporal lobe epilepsy by stereotactic technique. The behavior and EEG changes of the rats were observed. The extent of learning and memory impairment was evaluated by Morris water maze. After that, the rats were killed to observe pathologic changes of hippocampus and cortex by pathological examination. Results: Rats injected of kainic acid had temporal lobe epilepsy and different extent of learning and memory impairment. The escape latency of the epileptic rats in place navigation test prolonged, and the percentage of swimming time in platform quadrant of the epileptic rats decreased in spatial probe test, there was a significant difference from the control group (P〈0. 05). The decrease was more severe and came to the climax two months after seizure as time prolonged. Pathological changes showed that the degeneration and necrosis of pyramid cell aggravated gradually from area CAn to CA4, CA2, and CA1 in injected lateral hippocampus, then such also occurred in contralateral hippocampus, but the degree was significantly lower than the injected lateral. The above pathological changes culminated two months after the injection. Conclusion ; There is a correlation between learning and memory impairment and the degree of degeneration and necrosis in hippocampal pathological changes.
