摘  要:目的观察丁酸钠(NaB)对人卵巢癌细胞生长和分化的影响。方法以人卵巢癌细胞(SKOV3)为实验对象,分对照组、不同浓度的丁酸钠组。采用MTT法检测NaB作用后细胞的生长抑制率,电镜下观察细胞的超微结构,流式细胞仪测定细胞周期变化,放射免疫法测定培养基中CA125的含量。半定量RT-PCR检测癌基因c-erbB-2mRNA水平的变化。结果丁酸钠4.5mm/L能明显抑制SKOV3细胞增殖,细胞形态呈明显的良性分化,细胞堆积于G1期,培养液中CA125分泌量及c-erbB-2mRNA水平均降低。结论丁酸钠对人卵巢癌细胞有明显的诱导分化作用,可能通过抑制癌基因c-erbB-2mRNA的表达实现。【Objective】 To study the effect of induced differentiation of sodium butyrate(NaB) on human ovarian carcinoma cell line(SKOV3). 【Methods】 SKOV3 cells were cultured in vitro and treated with NaB of different concentration for different time, the proliferation was studied with MTT test. Electron microscopy was performed to observe the changes of microstructure. The cell cycle was analyzed by flow cytomctry. CA125 level was detected by radioimmunoassay and the change of c-erbB-2mRNA level was determined by semiquantiative RT-PCR. 【Results】 NaB could significantly inhibit proliferation and induce morphodifferentiation of SKOV3 cells. In cytometry analysis, SKOV3 cells lines accumulated in G1 phase. The amount of CA125 in culture media and the relative expression levels of c-erbB2 mRNA decreased in comparing with that of the control group after treated with NaB. 【Conclusion】 NaB can induce differentiation of ovarian cancer SKOV3 cells, c-erbB2 may play an important role in the pro cession.
