摘 要:目的:探讨乙酰肝素酶的基因表达,对卵巢癌细胞转移侵袭能力的影响。方法:用Boyden小室体外侵袭实验比较两株卵巢癌细胞HO-8910和HO8910-PM的体外侵袭能力强弱;采用半定量逆转录聚合酶链反应(RT-PCR)和原位杂交检测细胞乙酰肝素酶mRNA的表达情况;用MTT观察细胞生长能力的变化。结果:MTT检测结果显示卵巢癌细胞HO8910-PM细胞生长速度较快,而HO-8910细胞生长速度较慢。RP-PCR提示及原位杂交结果提示乙酰肝素酶在高转移性HO8910-PM细胞中表达强于转移性HO-8910细胞。Boyden小室体外侵袭实验表明,卵巢癌细胞HO8910-PM体外侵袭力较强,其穿膜细胞数为20.1±0.312,而HO-8910细胞体外侵袭力较弱,其穿膜细胞数为11.3±0.024。两者差异具有显著性意义(P<0.05)。结论:乙酰肝素酶基因的表达,能促进瘤细胞的生长和转移能力。Objection: To investigate the effects of heparanase gene on the invasive ability of ovarian cancer cells. Methods: Boyden chamber in vitro invasion assay was used to detect the invasion capacity in vitro in two ovarian tumor cell lines HO-8910 and 8910-PM. The mRNA expression level of heparanase in cell lines HO-8910 and HO8910-PM was evaluated by semi-quantitative RP-PCR and ISH(in situ hybridization). MTT was used to observe the change of proliferation of the two ovarian tumor cell lines. Results: Boyden chamber in vitro invasion assay indicated that the mean invasion percentage of HO-8910 cells (11. 3±0. 024)was significantly lower than that of 8910-PM (20. 1±0. 312)(P〈0. 05). Compared with HO-8910 cells, HO8910-PM ceils expressed significantly higher levels of mRNA of heparanase by RT-PCR and ISH. The growth of HO8910-PM cell was quick, but HO-8910 ceils grew slowly. Conclusion: It is suggested that human heparanase gene can facilitate the growth and metastasis of ovarian cancer cell lines HO-8910 and HO8910-PM.
