摘 要:探讨人外周血及脐血来源树突状细胞(DC)的生物学特性及其对宫颈癌细胞的直接杀伤效应。自外周血及脐血分离获得单核细胞,体外经rhGM-CSF、rhIL-4和rhTNF-α诱导培养,获得DC,采用流式细胞仪(FCM)检测DC相关表型;MTT法检测人外周血及脐血来源DC对宫颈癌细胞HeLa及CaSki的体外直接杀伤效应。结果显示,人外周血及脐血来源单核细胞诱导培养第6天后具有典型DC形态,FCM结果显示高表达HLA-DR(分别为99.29%和98.14%)、CD86(分别为99.45%和90.92%)和CD209(分别为87.44%和83.14%),而CD14表达率较低(分别为0%和3.32%);抑瘤实验显示人外周血及脐血来源DC对宫颈癌细胞HeLa及CaSki均有明显杀伤效应。研究提示,人外周血及脐血来源DC对人宫颈癌细胞具有直接杀伤效应,两者具有相似的生物学特性,为DC在肿瘤免疫治疗中的作用提供了新思路。The biological characeristics and their direct effect of dendritic cells(DC) from human peripheral blood and cord blood were investigated, in which DC were induced in vitro by cultivation of the monuclear cells from human peripheral blood and cord blood in the presence of cytokines, such as rhGM-CSF, rhlL-4 and rhTNF-α, and the related phenotype of DC was determined by flow cytometery(FCM). Meanwhile, the direct cytotoxis effect of DC from human peripheral blood and cord blood upon the carcinoma cell lines HeLa and CaSki cells was detected by MTT assay. It was found that the induced cells from the human peripheral blood and cord blood showed typical morphology of dendritic cell after 6 days cultivation. The FCM analysis demonstrated the high expression of HLA DR(99.29% and 98. 14%), CD86 (99. 45% and 90. 92%) and CD209 (87.44 % and 83.14 %) on these cells, respectively, while the expression of CD14 was rather low(0% and 3.2% respectively). As demonstrated by the MTT assay the cytotoxic effect of the DC derived from human peripheral blood and cord blood upon the carcinoma cell lines Hel.a cells as well as CaSki cells with similar biological characteristics was obviously found. This experimental result provides a new idea in the immunothrapy for tumors.
