摘  要:背景与目的:胶质瘤是颅内常见恶性肿瘤,治疗效果一直不令人满意,本文探讨雷公藤红素治疗人脑胶质瘤的可行性。方法:建立BALB/c裸小鼠SHG-44胶质瘤同种移植动物模型,将34只荷瘤裸鼠随机分为5组,空白对照组、高、中、低剂量雷公藤红素作用组、阳性对照组,采用腹腔内注射给药方法。雷公藤红素按三种浓度4mg/kg、2mg/kg、1mg/kg分组给药,每日1次,每周5天;阳性对照顺铂组按2mg/kg给药,每周两次;空白对照组只喂养不做任何处理。共给药四周。全部荷SHG44胶质瘤裸鼠于给药后第28天断颈处死并剔出肿瘤,将肿瘤组织分块处置。定期观察肿瘤生长情况测量肿瘤体积,绘制肿瘤生长曲线并计算抑瘤率。结果:5组荷SHG44胶质瘤裸鼠的移植瘤体积(处死后测量)比较,差异显著(F=5.519,P=0.002);与空白对照组相比,雷公藤红素4mg/kg组、雷公藤红素2mg/kg组均能抑制肿瘤生长(P<0.05),其体积抑瘤率分别为35%、26.9%。结论:雷公藤红素能明显抑制荷SHG44胶质瘤裸鼠的移植瘤生长,并存在剂量依赖性。BACKGROUND & OBJECTIVE: The aim of this study is to investigate the effects of Celastrol on the growth of a human malignant glioma cell line SHG-44 in vivo. METHODS: Animal models were constructed on BALB/c nude mice with subcutaneously transplanted neoplasma of SHG-44 glioma. The nude mice bearing with SHG44 glioma were devided into five groups by random selection, which were treated without drugs or with different doses of Celastrol (4mg/kg, 2 mg/kg and lmg/kg, ip twice weekly for four weeks)or with Cisplatin(2 mg/kg, ip twice weekly for four weeks). The dimension of xenografts were measured and the living state of nude mice with SHG-44 glioma were observed. RESULTS: Celastrol at both 4mg/kg and 2mg/kg can inhibit the growth of xenografis in nude mice with SHG44 glioma (P 〈0.05), and the tumor volume was reduced by 35% and 26.9% in both of Celastrol treated groups respectively compared with the control. CONCLUSION: Celastrol can inhibit the growth of a human malignant glioma cell line SHG-44 in vivo significantly and dosage-dependently.
