广西地区汉族人群CD14基因启动子-159C/T多态性研究

摘　　要：目的了解CD14基因启动子-159C/T多态性各等位基因及基因型在广西地区汉族人群中的分布频率,比较其在不同种族间分布的差异。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测208例汉族人的CD14基因型,并结合文献对不同种族间的CD14基因型和等位基因的分布频率进行比较分析。结果在广西汉族人群中CC,CT和TT基因型频率分别为14.9%,54.3%和30.8%;等位基因C及T频率分别为42.1%和57.9%,CD14基因多态性分布在男女间差异均无显著性(P>0.05)。与德国人、挪威人、希腊人、捷克人、以色列人、意大利人比较,CD14基因多态性差异存在显著性(P<0.05)。结论在广西地区汉族人群中存在CD14基因启动子-159C/T多态性,其在不同种族间的分布可能存在差异,这种差异可能是导致一些疾病在不同种族间的发生、转归和预后不同的遗传因素之一。Objective To study the allele frequencies and genotype distribution of CD14 gene promoter -159C/T polymorphism in normal Hans population in Guangxi Province,and to analyze the distribution of CD14 polymorphism among different races. Methods CD14 genotypes were examined by polymerase chain reaction- restriction fragment length polymorphism（PCR-RFLP） method in 208 Hart individuals. The frequencies of allele and genotype of CD14 polymorphism were compared with other ethnics. Results The frequencies of CC, CT and Tr genotypes were found to be 14.9%, 54.3% and 30.8% ;the frequencies of C and T allele were found to be 42.1% and 57.9% in the Hart populations. CD14 gene polymorphisms had no significant difference between male and female （P〈0.05）. Compared with Germans, Norwegians, Greeks, Czechs, Israel, Italians populations, the CD14 polymorphism was significantly different（P 〈 0.05）. Conclusion CD14 genc promoter-159C/T polymorphism exits in normal Hans population of Hu Bei, and its distribution is significantly different among ethnics. These differences might be one of the genetic factors contributing to the different pathogenesis; progression and pmgnesis in some diseases among ethnics.
