β-七叶皂甙钠对大鼠脑出血后炎性损伤的保护作用的研究

摘  要:目的探讨β-七叶皂甙钠对脑出血后血浆炎性反应因子损伤的保护作用。方法实验大鼠随机分为假手术组,模型组,七叶皂甙钠组,采用胶原酶Ⅶ诱导大鼠脑出血模型,术后2h七叶皂甙钠组腹腔注射β-七叶皂甙钠(2.8mg/kg),另2组给予等量生理盐水,连续用药(1次/d),分别在给药3d和7d后取血,应用酶联免疫吸附法和放射免疫分析法测定血浆细胞间黏附分子-1(ICAM-1)、肿瘤坏死因子-α(TNF-α)含量。结果与假手术组比较,模型组血浆ICAM-1和TNF-α含量均明显升高(P<0.01),与模型组比较,七叶皂甙钠组血浆ICAM-1和TNF-α含量均明显下降(P<0.01,P<0.05)。结论β-七叶皂甙钠对脑出血后神经细胞损伤具有保护作用,其机制可能与减少ICAM-1和TNF-α的产生,抑制炎症反应有关。Objective To observe the effect of aescinate on inflammatory reactive factors: intercellular adhesion molecular-1 ( ICAM - 1 ) and tumor necrosis factor-α (TNF-α) content in plasma after intracerebral hemorrhage in rats and explore its protective mechanism. Methods Rats were randomly divided into six groups : sham-operative group, model group, aescinate group. The rat model of intracerebral hemorrhage was induced by collagenase Ⅶ. After 2 hours, β-aescinate was injected intraperitoneally with dose of 2. 8 mg/kg in aescinate group per day, saline was given in the other two groups per day. These rats were killed 3 and 7 days later to detect plasma ICAM-1 and TNF-α levels by fluorospectrophotometry and radioimmunoassay. Results ICAM-1 and TNF-α content in plasma were obviously higher ( P 〈 0. 01 ) in model group than those of sham group ( P 〈 0. 01 ). ICAM-1 and TNF-α content in plasma were obviously lower ( P 〈 0. 01, P 〈 0. 05 ) in aescinate group than those of model group. Conclusions α-aescinate has an evident protective effect on injured neuron after intracerebral hemorrhage in rats, which may be related to decreasing ICAM-1, TNF-α content in plasma and inhabiting inflammatory reaction.
