摘  要:目的探讨炭肾血液灌洗对破伤风毒素的清除能力,建立破伤风毒素的实验室检测方法。方法厌氧培养破伤风梭菌,制取破伤风毒素;将破伤风毒素静脉注入兔子体内以制备破伤风动物模型,发病后以炭肾行血液灌洗。采用本研究建立的SABC-ELISA法检测炭肾灌洗前后血液中破伤风毒素的含量变化。结果建立起了间接双抗体夹心ELISA法检测破伤风毒素的实验室方法,检测下限可达破伤风梭菌培养上清的最大稀释度1∶50000左右,灵敏度极高。使用炭肾灌洗血液后,实验兔血清中破伤风毒素的含量下降80%以上。结论炭肾灌洗能有效清除血液中的破伤风毒素。并建立起了检测破伤风毒素的实验室方法。Objective To investigate the eliminating power of Charcoal kidney for tetanospasmin out of blood. And to set up the laboratory test for measuring the tetanospasmin. Methods C. tetani were anaerobic cuhured to obtain the tetanospasmin. The tetanospasmin was injected intravenous into rabbits to reproduce the animal model of tetanus. After first attack of tetanus symptom, the animal model was mounted on Charcoal kidney for hemoperfusion. The blood samples were drawn before and after hemoperfusion for tetanospasmin measurement by SABC-ELISA, which was originally set up in this study. Results The laboratory test for measuring tetanospasmin was first set up with SABC-ELISA method, which shown a very sensitive detective capacity for tetanospasmin, the dilution limit can reachl:500O0.The concentration of tetanospasmin in serum of rabbits was significantly reduced more than 80% after hemoperfusion with Charcoal kidney. Conclusions hemoperfusion with Charcoal kidney can effectively get rid the tetanospasmin out of blood. A sensitive laboratory test for measuring the tetanospasmin was also set up originally.
